摘要
用HPLC法的两种不同的方式(正相和反相体系)进行维生素A的测定,反相体系采用Lichrospher 80C_(18)为固定相,甲醇—水(98∶2)为流动相。UV(325nm)或萤光(Ex 335 nm,Em 470 nm)检测器:正相体系采用Lichrosorb Si 60为固定相,异辛烷—异丙醇(90∶1)为流动相,UV(325nm)检测器进行测试。实验证明采用这两种方式均可获得满意的结果,回收率为95.3%~102.1%,变异系数为0.56%~1.24%。
In this paper two different modes (normal- and reversed-phase) of HPLC for determining the contents of vitamin A are described. Reversed-phase adsorption systems with Lichrospher 80 C-18 as the stationary phase and mixtures of methanol-water(98: 2) as mobile phases were employed, using detective means UV(325 nm) or fluorescence (Ex=335, Em=470) detector. Normal-phase adsorption system with lichrosorb Si 60 as the stationary phase and mixtures of isooctan-2-propanol(99: 1) as mobile phase were employed, using U, (325 nm) detector. The two modes of HPLC proved to be satisfactory. The recovery rates were 95.3%~102.1%, the coefficients of variation were 0.56%~1.24%.
