摘要
[目的]探讨抑癌基因PTEN和Caspase鄄3在胃原发恶性淋巴瘤发生发展过程中的作用。[方法]采用SABC免疫组化方法观察56例胃原发恶性淋巴瘤组织中抑癌基因PTEN编码产物和细胞凋亡关键酶Caspase鄄3表达情况,并对胃淋巴瘤病理生物学行为特征进行比较分析。[结果]56例胃原发恶性淋巴瘤中28例PTEN表达阳性,阳性率50.0%(28/56),较非癌胃黏膜表达(96.4%,27/28)明显下降(P<0.05)。PTEN表达与胃原发恶性淋巴瘤的浸润深度和淋巴结转移密切相关(P<0.05)。Caspase鄄3在胃原发恶性淋巴瘤组织中阳性率76.8%(43/56),显著低于浅层胃黏膜表达率96.4%(27/28,P<0.05)。PTEN基因编码蛋白表达与Caspase鄄3的表达呈正相关(P<0.05)。[结论]抑癌基因PTEN在胃原发恶性淋巴瘤的发生发展中,可能通过影响Caspase鄄3的活性而起重要作用。
To investigate the role of anti-oncogene PTEN and cell apoptosis key factor Caspase-3 in primary malignant gastric lymphoma. The expression of PTEN protein and Caspase-3 were detected by immunohistochemicals method SABC in 56 cases with primary malignant lymphoma. Positive expression PTEN and Caspase-3 in the primary malignant gastric lymphomas (50.0%, 28/56; 76.8%, 43/56) was significantly lower than that in the normal or superficial gastric mucosa (96.9%, 31/34; 96.4%, 27/28). The expression of PTEN in gastric lymphoma was correlated not only with depth of invasion and lymph node metastasis but also with the expression of Caspase-3. [Conclusion] The anti-oncogene PTEN may play an important role in the development and progression of malignant gastric lymphoma by influencing the activity of Caspase-3.
