摘要
目的探讨大肠癌组织中胃泌素 (gastrin ,GAS)、生长抑素 (somatostatin ,SS)与大肠癌细胞凋亡指数 (apoptosisindex ,AI)和凋亡调控基因 p5 3的相关性。 方法采用免疫组织化学SABC法和原位凋亡检测TUNEL法 ,检测 6 2例大肠癌组织中GAS、SS、p5 3及细胞凋亡的表达。 结果在大肠癌组织SS高表达组、中表达组的AI明显高于SS低表达组 (q高与低 =5 0 6 ,q中与低 =3 95 ,均P <0 0 1) ;而在GAS各表达组中的AI变化与此相反。p5 3阳性表达率在SS (GAS)低表达组、中表达组、高表达组三组间相比较存在着明显差别 (χ2 SS=10 16 9,χ2 GAS=7 833,均P <0 0 5 ) ,其中p5 3在GAS高表达组、中表达组的阳性表达率明显高于低表达组 (χ2 高与低 =3 85 9,χ2 中与低 =4 796 ,均P <0 0 5 ) ;p5 3在SS高表达组的阳性表达率明显低于低表达组 (χ2 =6 710 ,P <0 0 5 ) ;而 p5 3在SS中表达组表达的阳性表达率低于低表达组 ,但差异无显著性 (χ2 =2 180 ,P >0 0 5 )。GAS/SS积分比值变化与AI呈负相关 ,与 p5 3呈正相关。 结论GAS、SS对大肠癌细胞凋亡的调控可能与 p5 3基因突变有关。
Objective To explore the correlation between the expression of somatostatin (SS), gastrin (GAS), apoptosis index (AI),and p53 gene in colonic carcinoma. Methods The expression of GAS, SS, p53 and apoptosis cell were detected by immunohistochemistry (streptavidin-biotin-pero xidase complex, SABC) and in situ apoptosis detecting technic (TUNEL) . Results AI is higher in SS high and middle expression cases than in low expression cases(q=5.06,q=3.95,P<0.01), and vice versa for GAS expression. The positive expression rate of p53 was significantly different among SS(GAS) high, middle and low expression group (χ2 SS=10.169, χ2 GAS=7.833,P<0.05).The positive expression rate of p53 was higher in GAS high and middle expression cases than low expression(χ2=3.859,χ2=4.796, P<0.05); The positive expression rate of p53 was lower in SS high expression cases than in low expression ( χ2=6.710,P<0.05).The integral of GAS/SS was positively correlated with p53 and negatively correlated with AI. Conclusion The GAS?SS regulates and controls the cell apoptosis in large intestine carcinoma, which maybe related with p53 gene mutation .
