两种方法提取的DNA在脆性X综合征基因FMR-I检测中的比较

摘要 目的 探索一种更简便、更快速的DNA提取方法用于脆性X综合征的基因检测。方法 用经典的苯酚 -氯仿 -异戊醇提取的DNA和用饱和盐法提取的DNA用于脆性X综合征的基因检测并进行比较。结果 两种方法提取的DNA在纯度和产量上存在差异 ,但在脆性X综合征基因检测包括完成PCR和SouthernBlot杂交两种实验时结果均无差异。结论 用饱和盐法提取的DNA可用于脆性X综合征基因的检测 ,它比传统的苯酚 -氯仿 -异戊醇提取法更简便、快速和经济 ,可用于临床上对脆性X综合征的筛查。 Objective To establish a simple and rapid method of extraction of genomic DNA on detecting FMR-1 gene of the fragile X syndrome. Methods The genomic DNA from whole blood samples was extracted by two different methods: phenol-chloroform and saturated salt,and then was compared in the experiments of fragile X syndrome gene analysis. Results There were difference in DNA yields and quality between the two methods, but DNA extracted by both methods were suitable for detecting the FMR-1 gene of the fragile X syndrome, including PCR test and Southern Blot hybridization. Conclusion The DNA extraction by saturated salt can be used for screening the fragile X syndrome clinically.
