摘要 目的 探讨过氧物酶体增殖因子活化受体 (PPAR)γ配体对人肝癌细胞系生长的影响。方法 用细胞计数、[3 H ] 胸苷摄入和流式细胞术分别检测 15 脱氧 △12 ,14 前列腺素J2(15d PGJ2 )和曲列格酮 (TGZ)对人肝癌细胞系BEL 740 2细胞增殖及细胞周期的影响。结果 15d PGJ2和TGZ均以剂量依赖方式抑制细胞生长 ,减少S期细胞比值和增加G0 /G1期细胞比值 ,但对G2 /M期细胞比值无明显影响。结论  15d PGJ2和TGZ均可抑制BEL 740 2细胞增殖 ,这种抑制作用可能部分与PPARγ介导的G1细胞周期停滞有关。 Objective To study the effect of peroxisome proliferator activated receptor (PPAR) γ ligands on cell growth in human liver cancer cells.Methods The effects of 15-deoxy-12,14-prostaglandin J2 (15 d-PGJ2) and troglitazone (TGZ) on cell proliferation and cell cycle were examined by cytometry,[ 3H]-thymidine incorporation and flow cytometry in BEL-7402 liver cancer cells.Results Both 15 d-PGJ2 and TGZ suppressed cell growth in a dose-dependent manner.At concentrations of 10 and 30 μmol/L,15 d-PGJ2 or TGZ increased the proportion of G0/G1 cells and decreased that of S cells,but there was no significant change in the proportion of G2/M cells.Conclusion Both 15d-PGJ2 and TGZ inhibit cell proliferation of BEL-7402 liver cancer cells,which may be due to PPAR-γ-mediated G1 cell cycle arrest in part.
