摘要
大熊猫皮肤组织块在 2 0 0IU/mL胶原蛋白酶Ⅰ和 0 2 5 %胰蛋白酶中 ,分别在 37℃、4℃下进行酶解的 4个方案中 ,以在 2 0 0IU/mL胶原蛋白酶Ⅰ ,在 4℃下酶解 4 8~ 72h的酶解效果最好 ,活细胞的得率达到 4 6 0 4× 10 6/g 。
In the paper, we studied that custaneous tissue of giant panda was digested with collagenase-Ⅰ (200?IU/mL) or trypsin (0.25%) under 37?℃ or 4?℃. In the 4 treatments, the effect shows: the method of enzyme digestion which was treated with collagenase-Ⅰ (200?IU/mL) under 4℃ for 48~72?h was the best in all methods and the rate of living cells reached 4.604×106/g. In addition, the fibroblast cells was main cells in the test treated with collagenase-Ⅰ (200?IU/mL).
