非洲爪蟾TGF-β5和小鼠TRP-2联合免疫诱导抗原特异性CTLs 被引量:2

摘要 目的 研究非洲爪蟾转化生长因子 β5 (aTGF β5 )基因免疫是否能增强肿瘤治疗性DNA疫苗诱导抗原特异性细胞毒性T淋巴细胞的能力。方法 构建编码小鼠肿瘤抗原酪氨酸酶相关蛋白 2 (mTRP 2 )的真核表达质粒 ,与aTGF β5真核表达质粒联合免疫C5 6BL 6小鼠 ,采用标准51 Cr释放试验检测抗原特异性细胞毒性T淋巴细胞的诱导。结果 aTGF β5和mTRP 2联合免疫可在体外诱导H 2Kb 限制的mTRP 2 aa1 80 - 1 88特异性细胞毒性T淋巴细胞产生。结论 aTGF Objective To study the potential of immunization with Xenopus laevis transforming growth factor beta 5 to enhance the efficacy of DNA vaccine for tumor to induce antigen-specific cytotoxic T lymphocytes. Methods The mammalian expression plasmid encoding murine tyrosinase-related protein 2 was constructed and C57BL/6 mice were immunized with a combination of the plasmid encoding Xenopus laevis transforming growth factor beta 5 and the plasmid encoding murine tyrosinase-related protein 2. Induction of antigen-specific cytotoxic T lymphocytes was assessed in standard 51Cr release assays. Results Immunization with a combination of Xenopus laevis transforming growth factor beta 5 and murine tyrosinase-related protein 2 elicited antigen-specific cytotoxic T lymphocyte reponse in vivo. Conclusion The results suggest that immunization with Xenopus laevis transforming growth factor beta 5 is capable of improving the efficacy of DNA vaccine encoding murine tyrosinase-related protein 2 to induce specific cytotoxic T lymphocytes.
