摘要 目的 探讨人白血病细胞系血管内皮生长因子 (VEGF)及其Flt 1受体的表达 ,进一步阐明VEGF在白血病细胞异常增殖中的作用机制。方法 以人脐静脉血管内皮细胞系ECV3 0 4作为对照 ,采用RT PCR半定量技术和免疫组化检测体外培养的人白血病细胞系K5 62和HL 60中VEGF及Flt 1的表达。采用MTT试验观察VEGF反义寡核苷酸 (VEGF AS)对白血病细胞体外增殖的影响。结果 K5 62和HL 60细胞同时表达VEGF和Flt 1mRNA和蛋白。VEGF AS能够抑制白血病细胞体外增殖。结论 在白血病细胞异常增殖中自分泌VEGF可能起着重要作用。 Objective To explore the expressions of vascular endothelial growth factor (VEGF) and its receptor Flt 1 in human leukemic cell lines (HL 60 and K562 cells) cultured in vitro and to clarify the effect of VEGF on the abnormal proliferation of leukemic cells. Methods Reverse transcription polymerase chain reaction (RT PCR) and immunohistochemistry were used to examine the expressions of mRNA and protein for both VEGF and Flt 1 in the two human leukemic cell lines, taking ECV304 (human umbilical vein endothelial cells) as the control cells. MTT test was used to examine the effect of VEGF antisense oligodeoxynucleotide (VEGF AS) on the proliferation of leukemic cells. Results The expressions of mRNA and protein of both VEGF and Flt 1 were found in the two human leukemic cell lines. VEGF AS could inhibit the proliferation of human leukemic cell lines in vitro . Conclusion The autocrine VEGF may play an important role in the abnormal proliferation of leukemic cells.
