Contents of Transforming Growth Factor-β_1 in Vivo for the Diabetic Nephropathy: Determination and Clinical Significance

摘要 目的探讨糖尿病肾病 (DN)患者体内转化生长因子 - β1(TGF - β1)含量及其与糖尿病微血管病变的关系。方法采用ELISA双抗夹心法对 4 8例 2型糖尿病肾病 (G1组 )患者尿液、血液及外周血单个核细胞PHA体外刺激培养上清液 (CPBMCs)中TGF - β1含量进行检测 ,并与 4 6例肾功能正常的 2型糖尿病 (G2组 )及 4 8例健康人(G3组 )比较。结果G1组尿液和血浆中的TGF - β1含量明显升高 ,其尿液中含量 (45 0 2 9± 99 75fg·L-1)与G2组 (74 72± 18 5 7fg·L-1)和G3组 (5 9 90± 9 5 5fg·L-1)比较 ,分别为P <0 0 1和P <0 0 0 1。血浆中含量 (387 4 5± 82 0 6fg·L-1)与G2组 (5 8 91± 11 0 3fg·L-1)和G3组 (47 2 5± 6 2 2fg·L-1)比较 ,分别为P <0 0 5和P <0 0 1。G2组与G3组之间比较 ,尿液及血浆中含量均无明显差异 (P均 >0 0 5 )。G1组患者CPBMCs中的TGF - β1含量与G2组、G3组之间无明显差异 (P均 >0 0 5 )。结论尿液和血浆中TGF - β1含量检测可能作为判断DN病情活动的一项有意义指标。 Objective To study and determine the contents of transforming growth factor-β 1(TGF-β 1)in diabetic patients with nephropathy,and the relationship between TGF-β 1 and diabetic microvascular complications.Methods TGF-β 1 level in urine,plasma and the culture supernatant of PHA mitogen-stimulated peripheral blood mononuclear cell(CPBMCs) in vitro were measured by ELISA,the samples of which were collected from 48 cases of type 2 diabetic patients with nephropathy(groups 1,G1),compared with 46 type 2 diabetic patients with normal renal function(G2) and 48 cases of healthy adults(G3) Results The level of TGF-β 1 in urine(450 29±99 75fg·L -1)and plasma(387 45±82 06fg·L -1) of G1 were all significantly higher than those of the control group.Conclusion TGF-β 1 contents of urine and plasma may be related to the severity of nephropathy and microvascular complications in diabetic patients.
