摘要
嗜热真菌ThermomyceslanuginosusCBS2 88 5 4 M1 8耐热木聚糖酶的产酶条件和酶谱分析结果表明 :玉米芯水不溶木聚糖相对于其它来源木聚糖为最佳碳源 ,而酵母提取物和蛋白胨作为复合氮源时效果最好。培养基最适初始pH值为 7 0 ,最适培养温度为 5 0℃。在最适条件下发酵所产木聚糖酶活力最高达 1 ,83 4u mL。另外 ,SDS PAGE和酶谱分析 (变性和非变性状态下 )结果都表明该菌只产生一种分子量约为 2 6kD的G 1
Cultivation conditions of the thermophilic fungus Thermomyces lanuginosus CBS288.54-M18 for the production of thermostable xylanase were investigated. Corn cob xylan (water-insoluble) was found to be the best inducer and C source. Tryptone+yeast extract was the best nitrogen sources. The optimal temperature and the initial pH of the cultures for xylanase production were 50 ℃ and pH 7.0, respectively. The highest xylanase activity of 1,834 u/mL was achieved under these optimal conditions, whereas the crude xylanase was stable for more than 6 hours at 60 ℃. In addition, SDS-PAGE and zymography analysis of culture supernatant under native and denature conditions revealed the presence of a single 26 kD xylanase of family G/11.
