神经节苷脂促进异体周围神经再生的实验观察 被引量:7

摘要 目的 观察神经节苷脂GM 1对异体移植周围神经再生的影响。方法 选择成年大白兔 36只。 4只作为供体 ,切取双侧坐骨神经 ,制成 12mm神经段 ,深低温冷冻储存 2周。其余 32只随机分为实验组和对照组。将兔左侧坐骨神经造成10mm长缺损 ,用复温后的异体神经进行移植桥接。然后作以下处理 :实验组局部应用GM1(1mg/d× 14 ) ,对照组不做任何处理。术后 12周进行组织学、电生理、形态学定量分析以及免疫学等观察。结果 无急性排斥反应发生。GM1组与空白对照组神经传导速度及形态学定量分析有显著性差异 (P <0 0 5 )。结论 外源性神经节苷脂GM1可促进异体移植周围神经再生。 Objective To study the effects of ganglioside (GM1) on the regeneration of peripheral nerve after allografts in rabbits.Methods Thirty-six heath adult New Zealand white rabbits were used in this study. Four rabbits were selected as donors. Bilateral sciatic nerves were taken out from them and cryopreserved in a -86℃ refrigerator for 2 weeks. The other thirty-two rabbits as recipients were divided into two groups (experiment and control) randomly. 10mm defect was created in left sciatic nerve , the experiment and control of the nerve allograft for bridging the defect of peripheral nerves were done after rapid thawing.The rest managements were as follow:(1)The experiment: local injecting GM 1 (1mg/d×14).The control:not any management;(2)Regeneration of the sciatic nerve in both groups were examined electrophysiologically, hislologically, and immunologically.Results The acute reject reaction was not found.analysis between the two groups was significant in 12 weeks after operation(P<0.05). Nerve regeneration in the experiment group was superior to that in the control group.Conclusion The results reveal that ganglioside stimulated the regeneration of allograft peripheral nerves.
