摘要
目的 探讨GES 1细胞体外培养的增殖及O2 ·的生成情况及PMA、DPI、SOD对GES 1生成O2 ·和GES 1增殖的影响。方法 体外培养GES 1细胞分别为 2 4、4 8、72h ,3 H Tdr掺入法测定GES 1细胞CPM值 ,了解GES 1细胞增殖情况。细胞色素C还原法测定GES 1细胞产生O2 · 水平 ,并观察PMA、DPI对GES 1生成O2 ·和GES 1增殖的影响 ,不同浓度SOD对GES 1增殖的影响。结果 GES 1细胞能够自身产生O2 ·,PMA增加GES 1细胞产生O2 ·,可以促进GES 1细胞的增殖。DPI和SOD减少GES 1细胞产生O2 ·,可以抑制GES 1细胞的增殖。结论 低水平增加O2 ·可以促进GES 1细胞的增殖。提示氧化 抗氧化作用可能在胃相关疾病的防治中具有一定的意义。
Objective To study the role of superoxide anion(O 2 -· )in promoting proliferation of GES-1 cell line.Methods GES-1 cells cultured were stimulated directly by phorbol 12-myristate 13-acetate (PMA),diphenylene iodonium(DPI),superoxide dismutase(SOD).The effect of O 2 -· in promoting proliferation of GES-1 cells was investigated by using 3H-Tdr incorporation liquid scintillation counter.The concentration of O 2 -· generated by GES-1 cells was measured by ferricytochrome C reduction.Results Proliferation of GES-1 cells was induced obviously by low concentration of O 2 -· .PMA promoted the proliferation of GES-1 cells by stimulating the generation of O 2 -· .DPI,SOD inhibited the proliferation of GES-1 cells by decreasing the generation of O 2 -· .Conclusions The effects of low concentration in promoting proliferation of GES-1 cells indicate the important role of oxidation and anti-oxidation in prevention and cure of some stomach diseases.The biological effects of O 2 -· was related closely with its dose.
