摘要
目的 探讨生长抑素类似物奥曲肽对人肝癌细胞株MHCC97 L中血管内皮生长因子 (VEGF)mRNA表达及VEGF合成与分泌的影响。方法 利用酶联免疫吸附法、流式细胞仪、逆转录 聚合酶链反应检测不同剂量的奥曲肽对人肝癌细胞株MHCC97 L的VEGF合成与分泌及VEGFmRNA表达的影响。结果 奥曲肽 (10 -5~ 10 -10 mol/L)可以抑制人肝癌细胞株MHCC97 L中VEGFmRNA表达及VEGF合成与分泌 ,呈剂量依赖性 ,可见饱和现象。结论 奥曲肽能够抑制人肝癌细胞株MHCC97 L中VEGF的表达、合成和分泌 ,可能是奥曲肽抗血管新生的作用机制之一。
Objective To investigate the effects of somatostatin analogue octreotide on vascular endothelial growth factor (VEGF) expression,production,and secretion by hepatocellular carcinoma (HCC) cell line MHCC97-L. Methods The effects of octreotide on VEGF mRNA expression,and intracellular protein production and secretion in HCC cell line MHCC97-L were measured by RT-PCR,flow cytometry and enzyme linked immunosorbant assay (ELISA). Results Octreotide in concentrations from 10 -5 mol/L to 10 -10 mol/L inhibited VEGF mRNA expression,intracellular production and secretion of VEGF in HCC cell line MHCC97-L in a dose-dependent manner. Conclusion Octreotide is a potent inhibitor of VEGF expression and production by HCC cell line MHCC97-L,suggesting that the inhibitory influence of octreotide on VEGF expression and production may be one of the mechanisms underlying its anti-angiogenesis effect on HCC.
