摘要
目的探讨Neurturin(NRTN)和神经生长因子 (nervegrowthfactor ,NGF)联合治疗对AD鼠学习记忆和海马突触素表达的影响。方法切断成年SD大鼠左侧穹窿海马伞 (Fimbria Fornix ,FF) ,建立AD鼠模型。治疗组左侧脑室注射NRTN、NGF ,用Y型迷宫检测大鼠学习记忆能力、免疫组化方法和图象分析技术对大鼠海马突触素进行定量分析。结果与NGF组、损伤组相比 ,联合组学会Y型迷宫所需训练次数减少 ( 5 9.2 0± 3 .60vs 98.40± 4.5 1)和正确反应次数增加 ( 9.0 0± 0 .40vs 3 .82± 0 .64 ) ,但未完全达到正常水平 ;联合组海马CA1区多形层、辐射层、腔隙分子层和齿状回分子层突触素免疫反应物的COD值均明显增高 ,差异有高度显著性。结论NRTN和NGF联合治疗能促进AD鼠学习记忆恢复和海马突触素的表达 ,效果较单用NGF治疗好。
ObjectiveTo study the effect of neurturin(NRTN) and nerve growth factor(NGF) on the ability of learning and memory and expression of synaptophysin in hippocampus of rats with fimbria-fornix(FF) transection. MethodsAdult SD rats were subjected to left FF transection and received continuous infusion of NRTN/NGF or NGF into the lateral ventricle. After four weeks,the learning and memory ability were measured by Y-maze test and the brain of rats were processed for synaptophysin immunocytochemistry. In the present study, we used computer-assisted optical density measurements of synaptophysin immunolabel. ResultsBy comparison with lesion and NGF group, the ability of learning(59.20±3.60 vs 98.40±4.51) and memory(9.00±0.40 vs 3.82±0.64) of NRTN+NGF group increased significantly , the COD value of SYH immunolabel of the polymorphous(MPM), radiatun, lacunosum and molecular lay of dentate gyrus(MDG) in NRTN+NGF group in creased significantly . ConclusionTreatmenting with NRTN and NGF simultaneously can improve the ability of learning and memory and expression of synaptophysin in hippocampus of rats with FF transection, comparison with treatment by single NGF the effect is better.
