摘要 目的:研究来氟米特活性代谢产物A771726对角质形成细胞分泌α-肿瘤坏死因子TNF-α,白介素IL-6及IL-8的影响。方法:以体外培养的角质形成细胞株HaCaT为对象,以不同浓度A771726处理正常及TNF-α刺激后细胞,用生物活性法及ELISA方法测定培养上清中TNF-α、IL-6及IL-8的分泌变化情况。结果:正常情况下,HaCaT细胞可自发分泌TNF-α、IL-6和IL-8;药物作用后分泌水平下降;TNF-α诱导下,HaCaT细胞IL-6及IL-8表达量均增加,加入来氟米特后则表达量均受到抑制。结论:来氟米特可抑制角质形成细胞分泌TNF-α、IL-6及IL-8。 Objective: To observe the effects of leflunomide on the secretion of TNF-α,IL-6 and IL-8 in HaCaT cell. Methods: HaCaT cell was cultured in vitro, and treated with different concentrations of leflunmide. Concentrations of IL-6 and IL-8 were determined by ELISA. Results: spontaneously. After treatment with leflunmide, the secretion levels of TNF-α, IL-6 and IL-8 decreased. The levels of IL-6 and IL-8 increased after induction of TNF-α, and decreased after leflunmide treatment. Conclusions: Leflunmide can inhibit the secretion of TNF-α,IL-6 and IL-8 in HaCaT cell.
