摘要
对 4 0 2株不同种属的酵母菌株进行了出筛、复筛、单倍体分离、诱变 ,从亲株Y 6 16 18(amet) (Saccharomycescerevisiae)和Y 378 4 15 (αleu) (Sacharomyceskluyveri)的杂交菌株中选育到一株富锌酵母菌株 (编号为ZGH 374 )。并初步优化了发酵条件 :培养基为 80g L糖浓度的麦芽汁、10g L蛋白胨、锌添加量 4 0 0 μg mL ,pH 6 .0 ,装液量4 0mL 2 5 0mL三角瓶 ,接种量 10 % (V V) ,培养起始添加锌盐 ,培养时间 30h。在优化的条件下 ,杂交菌株ZGH 374的生物量 (细胞干重 )达到 14 .3g L ,细胞锌含量可达到 9.3mg g ,锌总含量达到了 133mg L。
High-biomass, zinc-enriched yeast strain (No ZGH-374) was constructed by primary screening, isolation of haploid, mutagenesis and hybridization between the parental strains Y-6-16-18 (a met) (Saccharomyces cerevisiae)and Y-378-4-15 (α leu) (Sacharomyces kluyveri). The primary optimization of cultivation conditions was selected through fermentation tests. The highest biomass and Zn content of the strain was obtained 30h at 28℃ and 220r/min, when 40mL of the culture in 250-mL shake flasks was grown in wert containing 80g/L sugar, 10g/L peptone and 400μg/mL Zn 2+ . The initial pH was adjusted to 6.0. The inoculums volume was 10% (V/V). Under the optimized cultivation conditions, the biomass (dry weight) reached 14.3g/L, the Zn 2+ content reached 9.3mg/g and the total Zn 2+ reached 133mg/L.
