鹦鹉热衣原体重组主要外膜蛋白免疫肉鸡效果观察 被引量:11

摘要 利用鹦鹉热衣原体重组主要外膜蛋白 (MOMP)在大肠杆菌中的表达产物 ,纯化后与白油佐剂乳化成疫苗。对 3批疫苗物理性状、无菌试验、安全试验、最小保护剂量、效力检验、免疫期、保存期和田间试验等各项技术指标进行了测定。重组MOMP疫苗为乳白色油包水型疫苗 ,黏度为 8s·0 4mL-1;细菌培养阴性 ;10d肉鸡分别以5 0和 10 0 μg注射免疫后 ,观察 14d ,免疫鸡群除一过性发蔫外 ,临床采食、饮水 ,注射部位剖检无异常变化 ;以 2 5 ,5 0 ,10 0和 2 0 0 μg分别免疫肉鸡后 ,30d龄攻毒 ,发现 5 0 μg免疫组可以抵抗病原的攻击 ;选择每只 5 0 μg免疫肉鸡 ,分别在 4 0 ,5 0和 6 0d攻毒 ,其保护率分别为 94 % ,95 %和 84 %。疫苗 2~ 8℃可储存 2 4 0d ,室温 ( 2 1~ 2 5℃ )、37℃不要超过 30d ;利用本疫苗免疫肉鸡 4 0 0 0 0只 ,保护率达 93% ,对照组为 85 %。本疫苗安全性高、保护率高、无副作用 ,能有效控制鹦鹉热衣原体的侵害。 The purified recombinant major outer-membrane protein (MOMP) of Chlamydia psittaci(Cps)expressed in E. coli was mixed with oil adjuvant and manufactured as MOMP-vaccine. The various subjects of three batch vaccines were examined such as physical character, safety test, minimum effective dosage, effect test, immune period, stability test and field test. The viscosity of MOMP-vaccine was 8?s·0.4?mL -1 ; The MOMP and vaccine were free of bacteria in cultivating test. When the broilers were vaccinated by 50 and 100?μg per chicken on day 10, and observed everyday for two weeks, the chickens showed normal feeding and drinking except for low spirit for a short time; Using 25,50,100 and 200?μg per chicken respectively, it was found that 50?μg group could obtained ideal protection by challenging of a virulent strain on day 30. We also found that if the broilers were vaccinated by 50?μg vaccine, and then challenged with virulent strain on day 40,50 and 60,the protective rate were 94%,95% and 84%, respectively. The vaccine could be stored at 2~8?℃ for more than 240 days, while the immune protection period could maintained 30 days at room temperature or 37?℃. It was tested in field with 40?000 broilers, the treated groups acquired 93% protection, while the control groups only obtained 85% protection. All these results demonstrated that the MOMP-vaccine was safety; no adverse effect, highly protection, and it would play a role in the prevention of Chlamydia psittaci from disease.
