摘要
研究强心合剂(主药:鹿角、淫羊蒮等)治疗充血性心力衰竭(CHF)的作用机制。建立腹主动脉绪窄所致后负荷过量的大鼠CHF模型,并予强心合剂治疗。5周后,分别用TUNEL法检测心肌细胞凋亡指数,RT-PCR法检测心肌细胞凋亡基因Bcl-2和Bax mRNA的表达。同时测量CHF大鼠左室重量指数(LVMI),并分别以地高辛、卡托普利作为对照组进行比较。结果:本方不仅降低LVMI,还能促进Bct-2mRNA表达,抑制Bax mRNA表达,明显抑制心肌细胞凋亡。其疗效与卡托普利相当。结论:强心合剂可能通过调控凋亡基因而抑制心肌细胞凋亡,从而延缓或部分逆转心脏重构的发生、发展。
To study the possible mechanism of'Qiangxin Mixture 'in the treatment of congestive heart failure (CHF) ,the left ventricular mass index ( LVMI) , apoptotic index, expression of Bcl-2 and Bax mRNA by in situ Tdt-mediated dUTP nick end labeling(TUNEL) and RT-PCR were observed in the CHF rats by abdominal aortic constriction with special silver clips. Results:The Mixture could decrease LVMI and regulate the expression of Bcl-2 and Bax mRNA and inhibit the apoptosis of cardiac myocytes,with significant differences from the Digoxin group(P <0. 01) and no difference from the Captopril group. The Mixture is effective in delaying or reversing the cardiac remodeling possibly by regulating the expression of apoptotic genes and inhibiting apoptosis in cardiac myocytes.
