摘要
目的 探究人脐血血清对人骨髓基质细胞增殖的作用。方法 无菌条件下抽取新鲜脐血 ,经肝素抗凝、离心分离血清、灭活补体、微孔滤膜过滤制备人脐血血清 ,细胞按照一定浓度接种于DMEM / 2 0 %人脐血血清培养基 ,定期观察细胞生长并更换培养基。结果 细胞接种 2 4h后有少量梭形细胞贴壁 ,2周时细胞基本长满瓶底 ,细胞呈漩涡状排列。
Objective: To explore the effect of human umbilical cord blood serum on human bone marrow stromal cell proliferation.Methods: Fresh umbilical cord blood was collected sterily. After anticoagulating with heparin, centrifugalizing serum, inactivating complement, filtering serum with micropore, human umbilical cord blood serum was produced. Cells with certainly density suspended in culture medium DMEM containing 20% human umbilical cord blood serum, regularly observed cell proliferation and changed culture medium. Results: After on culture 24 hours some adherent fusiform cells were shown after; culture two weeks fusiform cells were distributed over the base of vessel, showing whirlpool arrangement. Conclusion: Human bone marrow stromal cells can proliferate in vitro in DMEM containing human umbilical cord blood serum.
