摘要
目的 :研究极低密度脂蛋白 (VL DL)对 SD大鼠系膜细胞 (MCs)的促增殖作用。 方法 :采用 MTT法测定不同浓度VL DL 在不同时间对 MCs的促增殖作用 ,采用流式细胞术观察 VL DL 诱发 MCs凋亡的情况。 结果 :不同浓度 VL DL(1 0~5 0 0 μg/ m l)在不同时间 (6~ 4 8h)与 MTT光密度值呈线性关系 ,r分别为 0 .91 1 (P<0 .0 1 )、0 .6 5 1 (P<0 .0 5 )。当 VL DL 浓度达 1 0 0 0 μg/ ml时 ,MCs出现凋亡。结论 :在一定浓度内 VL DL 可以时间剂量依赖性地促进 MCs的增殖 。
Objective:To study the proliferation promotion effect of very low density lipoprotein(VLDL) on mesangial cells(MCs) of SD rats. Methods:MTT method was used to measure the proliferation promotion effect of VLDL with different concentrations and treatment times. Flow cytometry was employed to observe the apoptosis of MCs for studying the toxicity of VLDL. Results:The optical density of MTT was linearly correlated with VLDL concentration(10 500 μg/ml)and treatment tims(6 48 h),with r being 0.911( P <0.01) and 0.651( P <0.05) respectively.When VLDL concentration reached 1 000 μg/ml, apoptosis occurred in MCs. Conclusion:VLDL can promote MCs proliferation in a time and dose dependent manner, while high concentration VLDL may promote MCs apoptosis due to cellular toxicity.
