摘要
芹菜(Apium graveolens L.)外值体能被含Ti质较载体的根癌农杆菌感染,目前已得到转基因的愈伤组织.该载体带有一个嵌合的npt-Ⅱ(新霉素磷酸转移酶Ⅱ)基因和一个胭脂碱合成酶基因.经胭脂碱测定和DNA分子杂交实验表明,外源的卡那霉素抗性基因已导入芹菜细胞.
A simple system for the transformation of Aprium graveolens L. has been developed. Cotyledon and hypocotyl explants from Aprium graveolens L. were cocultured with Agro-bacterium tumefaciens harboring a non-oncogenic Ti plasmidderired vector which contained a chi-maeric npt-II gene and a wild nopaline synthase gene. Transgenic calli had been obtained. Successful genetic transformation was confirmed by nopaline assay, NPT- I enzyme activity assay and DNA hybridization.
