电针对局灶性脑缺血大鼠神经细胞凋亡及神经生长因子的影响(英文) 被引量:14

摘要 目的:探讨电针督脉经大椎、百会穴对大鼠缺血区脑组织神经细胞凋亡及神经生长因子(nervegrowthfactor,NGF)的影响,为临床针灸治疗缺血性脑血管疾病奠定理论基础。方法:选用健康雄性Wistar大鼠24只。按随机数字表法分为假手术组、缺血组、缺血+电针组,每组8只。用凝闭一侧大鼠大脑中动脉的脑缺血大鼠为动物模型,采用TUNEL染色法和免疫组化染色S0-P法进行观察。结果:缺血组及缺血+电针组中每个脑片1000个神经细胞中凋亡细胞数目分别为676±12及326±15,缺血+电针组中,大脑皮质梗死区内TUNEL染色阳性细胞较缺血组显著减少(P<0.01);缺血+电针组中NGF受体免疫阳性表达在细胞数量上及强度上均较缺血组及假手术组明显增强(P<0.01)。结论:电针能抑制脑缺血后脑内神经细胞凋亡,可增强局灶性脑缺血大鼠脑组织的NGF受体的表达,对缺血性脑损伤具有一定的保护作用。 AIM:To study the influence of electroacupuncture for dazhui(GV 14) and baihui(GV 20) on apoptosis and nerve growth factor(NGF) of cerebral ischemic rats so as to provide the theoretical basis for acupuncture in the clinical treatment of ischemic cerebrovascular disease.METHODS:Totally 24 healthy male Wistar rats were randomized into pseudo-operation group,ischemia group,and ischemia+electroacupuncture group,with 8 in each,according to numerical method.Their one side middle cerebral arteries were obstructed and observed with TUNEL staining and immunohistochemical S-P staining.RESULTS:In the ischemia and ischemia+electroacupuncture groups,the cells of apoptosis 676±12 and 326±15 respectively in every 1 000 neurocytes.In the ischemia+electroacupuncture group,the number of TUNEL positive cells in the infarction area of cerebral cortex were obviously less than that of the ischemia group(P< 0.01). In the ischemia+electroacupuncture group,the quantity and intensity of NGFR immune positive expression were clearly increased than those in the other two groups (P< 0.01). CONCLUSION:Electroacupuncture functions to inhibit apoptosis,strengthen NGFR expression,and protect the cerebral tissue in ischemia.
