摘要
目的 :研究姜黄素 (Curcumin)对人胆管癌细胞系QBC939的生长抑制与促凋亡作用。方法 :采用MTT法检测姜黄素对胆管癌细胞的抑制作用 ;相差显微镜 ,电镜下观察细胞形态变化 ;琼脂糖凝胶电泳测定DNALadder ;流式细胞术分析DNA含量。结果 :1,2 ,4 ,8,和 16mg·L-1姜黄素均能抑制人胆管癌细胞QBC939的生长 ,且抑制率具有浓度和时间依赖性 ,IC50 为 5 6mg·L-1。 8mg·L-1姜黄素作用后 ,QBC939细胞出现典型的凋亡形态特征 ,DNALadder出现 ,并检测到亚二倍体凋亡峰。结论 :Curcumin能有效地抑制QBC939细胞生长并促进其凋亡。
Objective:To study the growth inhibited and apoptosis triggered effect of Curcumin on the human cholangiocarcinoma cell Line QBC939,Methods:Cell proliferation was assessed by MTT assays;Morphological changes of apoptosis were observed with invert microscope and transmission electron microsope.DNA Ladder and DNA content were examined by DNA agarose gel electronphores and PI fluorescence flow cytometry.Results:The cell growth was significantly inhibited by the different concentration (1,2,4,8,16mg·L -1 ) of Curcumin as revealed by cell counting and cell growth curve.The inhibitory rate was depend on concentration and treatment time.The IC 50 value was found to be 5 6mg·L -1 .QBC939 cell line treated by Curcumin in a concentration of 8mg·L -1 were typical of apoptotic morphology with aneuploid peak(AP),meanwhile,DNA Ladder were seen.Conclusion:The Curcumin not only inhibites proliferation but also induces apoptosis of human cholangiocarcinoma cell line QBC939.
