摘要
目的 从脑挫裂伤发生后血清一氧化氮、内皮素改变与海马区神经细胞凋亡之间的相关性入手 ,研究创伤性脑损伤后继发性颅脑损伤的分子生物学机制。方法 按照改良Feeney自由落体脑损伤模型方式制作大鼠左顶叶脑挫裂伤模型 ,采用硝酸还原酶法、酶联免疫吸附法、原位细胞凋亡检测技术分别对脑挫裂伤后共十个时相点的各指标进行检测 ,所得数据均以SAS( 6.12 )统计软件包进行配对t检验及多元线性相关分析。结果 血清一氧化氮、内皮素及海马区神经细胞凋亡分别在脑挫裂伤后不同时相升高 ,随后下降 ,前两者彼此之间以及前两者与后者间均表现为正相关。结论 大鼠脑挫裂伤后血清一氧化氮、内皮素的变化之间以及此两者与神经细胞凋亡之间存在着特定关系 ,前两者相互影响。
Objective The mechanism of molecular biology of secondary craniocerebral injury after traumatic cerebral injury was investigated by analyzing the relationship between the changes of nitric oxide,endothelin and the neuron apoptosis in hippocampal region.Methods The models of rat cerebral contusion were made according to the modified Feeney method.The related parameters in different times after cerebral contusion were detected by nitric acid reductase method,enzyme linked immunosorbent assay,in situ cell apoptosis detection (TUNEL) respectively.Results The levels of nitric oxide and endothelin in serum and the apoptosis positive neurons in the hippocampal region increased in different times after cerebral contusion and decreased afterwards,and the former two parameters were positively correlated to each other and the two parameters were positively correlated to the latter one.Conclusion After cerebral contusion,there may exist a specific relationship among serum nitric oxide,serum endothelin and neuron apoptosis in hippocampal region,and the former two parameters may have influence on each other,which results in neuron apoptosis.
