黑地黄丸对肾纤维化模型大鼠肾组织炎症因子及TLR2/NF-κB/Myd88通路的影响 被引量:11

Effects of Heidihuang Pills(黑地黄丸) on Renal Tissue Inflammatory Factors and TLR2/NF-κB/Myd88 Pathway in Renal Fibrosis Model Rats

摘要 目的观察黑地黄丸对肾纤维化大鼠肾组织微炎症状态的影响及可能作用机制。方法将50只Wistar大鼠随机分为正常组、模型组和黑地黄丸高、中、低剂量组,每组10只。除正常组外,其余各组大鼠均采用5/6肾切除法建立肾纤维化动物模型。从术后当日,黑地黄丸高、中、低剂量组大鼠分别给予含生药2、1、0. 5 g/ml药液按10 ml/kg灌胃,模型组与正常组给予同等体积生理盐水,每天1次。连续给药60天后,检测各组大鼠血清肌酐(Cr)、尿素氮(BUN)、超敏C反应蛋白(hs-CRP)水平,HE染色观察大鼠肾组织病理变化,免疫组化和Western blot法检测肾组织Toll样受体2 (TLR2)、核转录因子κB (NF-κB)、髓样分化因子88 (Myd88)蛋白表达,免疫组化法检测肾组织炎症因子肿瘤坏死因子(TNF-α)、白细胞介素6(IL-6)表达。结果与正常组比较,模型组大鼠血清Cr、BUN、hs-CRP水平明显升高,肾组织中TLR2、NF-κB、Myd88、TNF-α、IL-6表达亦升高(P <0. 05);与模型组比较,黑地黄丸高、中、低剂量组大鼠血清Cr、BUN、hs-CRP水平明显降低,肾组织中TLR2、NF-κB、Myd88、TNF-α、IL-6表达亦降低(P <0. 05);黑地黄丸高、中、低剂量组组间比较,各指标差异无统计学意义(P> 0. 05)。HE染色结果显示,模型组大鼠肾小球变形、肾小管间质增宽,炎性细胞浸润,而黑地黄丸高、中、低剂量组大鼠肾间质水肿、肾小球硬化、肾小管萎缩和炎性细胞浸润较模型组减轻,并且各剂量组之间差异不明显。结论黑地黄丸能明显改善肾纤维化大鼠肾组织微炎症状态,其作用机制可能与调节肾组织TLR2/NF-κB/Myd88通路有关。 Objective To observe the effects and its possible mechanism of Heidihuang Pills(黑地黄丸)on the microinflammation of renal tissue in rats with renal fibrosis.Methods A total of 50 Wistar rats were randomly divided into a normal group,a model group and a Heidihuang Pills high,middle and low dose groups,with 10 rats in each group.Except the normal group,the rats were performed 5/6 nephrectomy to establish a renal fibrosis model.From the day after surgery,rats in the high,middle and low dose groups of Heidihuang Pills were given intragastric administration of drug solution respectively containing 2,1,0.5 g/ml crude drug with 10 ml/kg,and the model group as well as the normal group were given the same volume of normal saline,once a day,continuous administration for60 days.The levels of serum creatinine(Cr),blood urea nitrogen(BUN)and high-sensitivity C-reactive protein(hs-CRP)were measured in each group.The pathological changes of renal tissue were observed by HE staining.Immunohistochemistry and western blot were used to detect the expression of Toll-like receptor 2(TLR2),nuclear factorκB(NF-κB)and myeloid differentiation factor 88(Myd88)in renal tissues.Immunohistochemistry was used to detect the expression of inflammatory factor including tumor necrosis factor(TNF-α),interleukin 6(IL-6)in renal tissue.Results Compared with the normal group,the levels of serum Cr,BUN and hs-CRP in the model group were significantly increased,and the expressions of TLR2,NF-κB,Myd88,TNF-αand IL-6 in renal tissues were also increased(P<0.05).Compared with the model group,the serum levels of Cr,BUN and hs-CRP in the high,middle and low dose of Heidihuang Pills groups were significantly decreased,and the expressions of TLR2,NF-κB,Myd88,TNF-αand IL-6 in renal tissues were also decreased(P<0.05).There was no significant difference in the indexes of the high,middle and low dose of Heidihuang Pills groups(P>0.05).HE staining showed that the glomerular deformation,tubulointerstitial widening and inflammatory cell infiltration in t
