红参对H2O2诱导H9c2大鼠心肌细胞氧化应激损伤的保护作用 被引量:14

摘要 目的探讨红参有效成分对心肌细胞氧化应激损伤的保护作用。方法过氧化氢(H2O2)诱导H9c2大鼠心肌细胞建立体外心肌细胞氧化应激损伤模型;MTT法检测细胞存活率;试剂盒检测细胞因子LDH释放、SOD活性和MDA含量;流式细胞术检测ROS、细胞凋亡;Western Blot法检测细胞凋亡相关蛋白表达。结果与模型组相比,红参有效成分预处理后,心肌细胞存活率显著提高,LDH水平下降,SOD活性显著提高,MDA水平受到抑制,ROS水平显著降低,细胞凋亡率显著降低。同时红参干预后心肌损伤细胞促凋亡蛋白Bax下调,抗凋亡蛋白Bcl-2表达上调,Bcl-2/Bax比值升高。结论红参通过降低ROS水平和细胞凋亡保护心肌细胞氧化应激损伤。 Objective To investigate the protective effect of active ingredients of red ginseng on oxidative stress injury of cardiomyocytes.Methods Hydrogen peroxide(H2O2)induced H9c2 rat cardiomyocytes to establish an oxidative stress injury model of cardiomyocytes in vitro.MTT assay for cell viability.Kit for detection of cytokine LDH release,SOD activity and MDA content.Flow cytometry ROS,apoptosis.Western Blot assay for apoptosisrelated protein expression.Results Compared with the model group,the survival rate of red ginseng was significantly increased,the LDH level was decreased,the SOD activity was significantly increased,the MDA level was inhibited,the ROS level was significantly decreased,and the apoptosis rate was significantly decreased.At the same time,after the intervention of red ginseng,the expression of Bcl-2 was down-regulated and the ratio of Bcl-2/Bax was upregulated,and the ratio of Bcl-2/Bax was increased.Conclusion Red ginseng protects cardiomyocytes from oxidative stress damage by reducing ROS levels and apoptosis.
