痛泻要方对肠易激综合征肝郁脾虚证大鼠中枢神经系统SERT的影响 被引量:21

Effect of Tongxie Yaofang on SERT of Central Nervous System in Rats with IBS Liver Depression and Spleen Deficiency Syndrome

摘要 目的:探讨痛泻要方治疗肝郁脾虚证大鼠,对中枢神经系统中海马、皮质、下丘脑5-羟色胺转运体的作用机制。方法:将120只SPF级SD大鼠随机分为6组:正常组、病理组、痛泻要方低剂量组、痛泻要方中剂量组、痛泻要方高剂量组、西药(匹维溴铵)组,每组20只。除正常组外,其余各组均用'束缚应激+番泻叶灌胃法'进行造模,每只大鼠前肢束缚2周,在束缚的基础上进行番泻叶灌胃1周,1次/d,造模结束后用相应剂量的痛泻要方和匹维溴铵进行灌胃治疗7 d。造模及治疗结束后,检测各组大鼠海马、皮质、下丘脑中5-羟色胺转运体的含量。结果:与正常组相比,模型组大鼠海马、皮质、下丘脑中5-羟色胺转运体含量明显降低(P<0.01);与模型组相比,痛泻要方低、中、高剂量组SERT含量回升。综合来看,以中、高剂量组升高效果最为明显(P<0.05或P<0.01);与模型组相比,西药组大鼠SERT含量变化没有统计学意义(P>0.05)。结论:痛泻要方治疗肠易激综合征肝郁脾虚证大鼠,可能与升高其海马、皮质、下丘脑中5-羟色胺转运体的含量有关。 Objective:To discuss the mechanism of Tongxie Yaofang on IBS of liver-depression and spleen-deficiencyrats’hippocampus,cortex and hypothalamus 5-HT transporter in their central nervous system.Method:A total of 120 SPF SD rats were divided into six groups(20 rats in each group):normal group,pathological group,low doses group of Tongxie Yaofang,middle doses group of Tongxie Yaofang,high doses group of Tongxie Yaofang and western medicine(Pinaverium Bromide)group.Each group made model by restraint stress and Fanxieye(Folium Sennae)by gavage except the normal one.Making models should constrain rats’fore limb for two weeks,and then Fanxieye(Folium Sennae)was given by gavage once a day for one week.Then we used the corresponding dose of Tongxie Yaofang or Pinaverium Bromide for seven days.Finally,we detected the contents of rats’hippocampus,cortex and hypothalamus 5-HT transporter.Result:Rats’hippocampus,cortex and hypothalamus 5-HT transporter of model group was significantly lower than that of the normal group(P<0.01)and the contents of SERT in different doses groups of Tongxie Yaofang were higher than those of the model group.The middle and high doses groups of Tongxie Yaofang were more effectively(P<0.05 or P<0.01),and the change of western group rats’contents of SERT made no statistically significantcompared with that of the model group(P>0.05).Conclusion:It may be relatedthat Tongxie Yaofang can make the contents of hippocampus,cortex and hypothalamus 5-HT transporter higher,and so it can treatliver-depressionand spleen-deficiency IBS rats.
