摘要
目的:优化金鉴灵软膏制备工艺,并对其治疗烧烫伤的作用进行初步药效研究。方法:选择姜黄素提取率为评价指标,以煎炸时间、煎炸温度、加油量、浸泡时间为影响因素进行单因素实验,在单因素分析的基础上增加皮肤愈合率为评价指标,Box-Behnken响应面优化制备工艺。采用深Ⅱ度烫伤模型,通过测定创面愈合率及愈合时间等指标,考察金鉴灵软膏对创面愈合情况的影响。同时通过ELISA法测定血清中TNF-α和IL-1水平,初步探讨其作用机制。结果:最优制备工艺为加麻油4倍量,浸泡23 h后,于144℃煎炸64 min;金鉴灵软膏组对于烫伤小鼠受损皮肤的愈合效果显著,血清TNF-α和IL-1水平均下降。结论:本实验优选的制备工艺稳定可行,为确保开发出安全有效、适合工业化生产的外用制剂提供了实验依据与技术支撑。
Objective:To optimize the preparation process of Jinjianling ointment and study its preliminary action in the treatment of burns.Methods:The extraction yield of curcumin was selected as the evaluation index.The single factor experiment was carried out with the frying time,frying temperature,fueling amount and soaking time as the influencing factors.The skin healing rate of the evaluation index was increased based on the single factor analysis.Box-Behnken response surface was used to optimize the preparation process.The deep II degree scald model was used to determine the effect of Jinjianling ointment on wound healing by measuring the wound healing rate and healing time.At the same time,the levels of TNF-αand IL-1 in serum were determined by ELISA,and the preliminary mechanism of action was investigated.Results:The optimal extraction process was adding 4 times of sesame oil,immersing for 23 hours,and frying at 144℃for 64 minutes.Jinjianling ointment group had significant healing effect on damaged skin of scalded mice,and both serum levels of TNF-αand IL-1 decreased.Conclusion:The optimal preparation process of Jinjianling ointment is stable and feasible,providing experimental basis and technical support for developing safe and effective preparation suitable for industrial production.
