摘要
为明确分离自茶卷叶蛾和中华大刀螳僵虫的2株虫生真菌的归属及生防潜力,采用形态学特征和r DNA-ITS序列分析其分类地位,测定了其对茶卷叶蛾和茶小卷叶蛾幼虫的致病力,并初步筛选了其最适培养条件。结果表明:2株真菌均为环链棒束孢Isaria cateniannulata,分别命名为ICBS918和ICTL911。在1.0×108孢子/m L浓度下,ICBS918和ICTL911对茶卷叶蛾幼虫的累计校正死亡率均达100%,LT50分别为3.13 d和3.15 d;对茶小卷叶蛾幼虫的累计校正死亡率分别为100%和95%,LT50分别为3.25 d和3.31 d。接菌后8 d,2菌株对茶卷叶蛾幼虫的LC50分别为0.47×105孢子/m L和1.01×105孢子/m L;对茶小卷叶蛾幼虫的LC50分别为2.20×105孢子/m L和1.34×105孢子/m L。2菌株23℃下最适生长培养基均为综合马铃薯培养基,最适产孢培养基均为蛋白胨马铃薯葡萄糖琼脂培养基;在萨氏培养基上,菌株ICBS918和ICTL911分别在24~30℃及24℃时生长速度最快,在27℃和21℃时产孢量最大。表明2株环链棒束孢对茶卷叶蛾和茶小卷叶蛾幼虫均具有较好的生防潜力,可作为生防菌进行开发和应用。
In order to confirm the taxonomic status and biocontrol potential of two isolates of entomopathogenic fungi,which were isolated from natural cadavers of Homona coffearia and Tenodera sinensis,morphological characters and molecular analysis of ITS-r DNA were carried out to identify their taxonomic position,the pathogenicity of the two isolates against the larvae of tea tortrix H. coffearia and smaller tea tortrix Adoxophyes honmai were measured by indoor bioassay,and the optimum culture conditions for the two isolates were preliminarily screened. The results showed that the two isolates were both identified as Isaria cateniannulata and named as ICBS918 and ICTL911,respectively. When H. coffearia were dealt with ICBS918 and ICTL911 suspension( 1. 0 × 108 spore / m L),the correctedaccumulative mortalities were both 100%; LT50 values were 3. 13 d and 3. 15 d,respectively. When A.honmai were treated under the same conditions,the corrected accumulative mortalities were 100% and95%,respectively; LT50 values were 3. 25 d and 3. 31 d,respectively. When H. coffearia were dealt with ICBS918 and ICTL911,the LC50values( on the 8th day) were 0. 47 × 105 spores / m L and 1. 01 × 105 spores / m L,respectively. When A. honmai were treated under the same conditions,the LC50 values were2. 20 × 105 spores / m L and 1. 34 × 105 spores / m L,respectively. The optimal culture media( 23 ℃) for mycelial growth of the two isolates was CPDA,and for sporulation PPDA. When ICBS918 and ICTL911 were cultured on SDAY medium,the optimal culture temperatures for mycelial growth were 24- 30 ℃and 24 ℃,and for sporulation 27 ℃ and 21 ℃,respectively. In conclusion,the two I. cateniannulata isolates have great potential for biocontrol of the larvae of H. coffearia and A. honmai.
