摘要
目的:探究哌拉西林钠(PS)对CCl_4诱导的大鼠急性肝损伤的作用及机制。方法:将40只大鼠随机分为对照组(Control,Ctrl)、PS组、四氯化碳(CCl_4)组和CCl_4+PS组,PS组和CCl_4+PS组每天腹腔注射PS(1 g/kg),连续给药7 d后,CCl_4组和CCl_4+PS组大鼠腹腔注射CCl_4溶液(1 ml/kg)。48 h后处死大鼠,试剂盒检测血清丙转氨酶(ALT)、谷草转氨酶(ALT)、白介素-6(IL-6)、IL-1β和IL-10的浓度,并检测肝组织超氧化物歧化酶(SOD)、谷胱甘肽(GSH)及丙二醛(MDA)的水平,Western blot检测Bcl-2和Bax表达,免疫组化检测凋亡蛋白Caspase-3的表达。结果:与Ctrl组比较,CCl_4组大鼠肝组织出现损伤,血清ALT和AST含量明显升高;与CCl_4组比较,CCl_4+PS组大鼠肝组织损伤减轻,ALT和AST浓度显著降低; CCl_4组大鼠血清炎症因子IL-6和IL-1β的浓度明显高于Ctrl组,IL-10浓度明显降低; CCl_4+PS组IL-6和IL-1β的浓度较CCl_4组比较明显降低,IL-10浓度明显升高;同时,CCl_4还能明显降低肝组织SOD和GSH浓度,升高MDA浓度,PS能显著升高模型大鼠SOD和GSH浓度,降低MDA浓度;此外,CCl_4能显著降低Bcl-2的表达水平及Bcl-2/Bax的比值,升高Bax和Caspase-3的表达水平; PS能显著减弱CCl_4对Bcl-2、Bax和Caspase-3的调控作用。结论:PS能通过抑制氧化应激减轻CCl_4诱导的大鼠急性肝损伤。
Objective: To investigate the effects and mechanisms of piperacillin sodium( PS) on CCl4-induced acute liver injury in rats. Methods: 40 rats were divided into Control( Ctrl),PS,CCl4 and CCl4+PS group. Rats in PS and CCl4+PS group were intraperitoneally injected with PS( 1 g/kg),and rats in CCl4 and CCl4+PS group were injected with CCl4 after rats treating with PS for 7 d. Rats were sacrificed and the concentrations of alanine transaminase( ALT),alanine transaminase( ALT),interleukin-6( IL-6),IL-1β and IL-10 in serum were measured by kits. And the levels of superoxide dismutase( SOD),glutathione( GSH),malondialdehyde( MDA) in liver tissue were detected by kits. The expressions of Bax and Bcl-2 were determined by Western blot. Immunohistochemistry was performed for level of Caspase-3. Results: Compared with Ctrl group,liver was injured and the concentrations of AST and ALT in serum were increased in CCl4 group. Compared with CCl4 group,the liver injury was alleviated and the concentrations of AST and ALT were decreased in CCl4+ PS group. The concentrations of inflammatory cytokines,including IL-6 and IL-1β were increased and IL-10 was decreased in CCl4 group compared with Ctrl group;but IL-6 and IL-1β were decreased and IL-10 was increased in CCl4+ PS group compared with CCl4 group. Meanwhile,CCl4 down-regulated the concentration of SOD and GSH but up-regulated MDA;PS increased the concentrations of SOD and GSH,but decreased MDA of model rats. In addition,CCl4 decreased the level of Bcl-2 and the ratio of Bcl-2/Bax,and increased the levels of Bax and Caspase-3 significantly;PS attenuated the effects of CCl4 on the expression of Bcl-2,Bax and Caspase-3. Conclusion: PS alleviated CCl4-induced acute liver injury of rats through inhibiting oxidative stress.
