摘要
目的探究补肾中药对去势大鼠雌性激素、骨代谢及破骨细胞活性的影响。方法选取雌性SD大鼠40只,建立骨质疏松模型,分为假手术组、模型组、补肾中药组、密钙息组,10只/组。各组对应给予生理盐水、补肾中药、密钙息注射液治疗3个月。检测各组大鼠血清钙(Ca)、磷(P)、碱性磷酸酶(ALP)含量;用ELISA法检测血清雌二醇(E2)、骨钙素(BGP)、抗酒石酸酸性磷酸酶(TRAP5b)、Ⅰ型胶原C端肽(CTX)、羟赖氨酸糖苷(GHYL)、肿瘤坏死因子-α(TNF-α)、白细胞介素-17(IL-17)、NF-κB受体激活蛋白配体(RANKL);检测腰椎骨密度(BMD)及最大承载力。结果与模型组比较,补肾中药组大鼠血清Ca、P下降,E2升高,骨形成指标ALP、BGP升高,胶原蛋白的分解产物CTX、GHYL升高,破骨细胞分泌的TRAP5b也升高,炎症因子TNF-α、IL-17、NF-κB水平均降低,腰椎BMD及最大承载力上升,差异均有统计学意义(P<0.05)。结论补肾中药能通过提升去势大鼠雌激素水平,改善骨代谢,增加骨量,抑制炎症反应及破骨细胞活性,达到防治骨质疏松的效果。
Objective To explore the effects of kidney-tonifying herbs on estrogen,bone metabolism and osteoclast activity in ovariectomized rats.Methods Forty female SD rats were selected to establish osteoporosis models,and were divided into sham operation group,model group,kidney-tonifying herbs group and miacalcic group,with 10 rats in each group.The four groups were treated with normal saline,kidney-tonifying herbs and miacalcic injection respectively for 3 months.Serum levels of calcium(Ca),phosphorus(P)and alkaline phosphatase(ALP)were measured in each group.Serum estrogen(E2),osteocalcin(BGP),tartrate-resistant acid phosphatase(TRAP5b),typeⅠcollagen C-terminal peptide(CTX),hydroxylysine glycoside(GHYL),tumor necrosis factor-α(TNF-α),interleukin-17(IL-17)and NF-κB receptor activator ligand(RANKL)were detected by ELISA.The lumbar spine bone mineral density(BMD)and maximum bearing capacity were detected.Results Compared with model group,serum Ca and P decreased and E2 increased,and the bone formation indexes ALP and BGP increased,and the collagen decomposition products CTX and GHYL increased,and TRAP5b secreted by osteoclasts also increased,and the inflammatory factors TNF-α,IL-17 and NF-κB decreased,and lumbar spine BMD and maximum carrying capacity increased,the difference was statistically significant(P<0.05).Conclusion Kidney-tonifying herbscan prevent and treat osteoporosis by raising estrogen level,improving bone metabolism,increasing bone mass,inhibiting inflammatory reaction and osteoclast activity in ovariectomized rats.
