摘要
目的 :选择一种高效、简便、稳定的从蜡块中提取基因组DNA进行聚合酶链反应 (PCR)扩增的方法。 方法 :分别采用四种不同的方法从石蜡组织中提取DNA ,比较所提取的DNA质量 ,PCR扩增cyclinD1基因的效率。对其中的一种方法进行了改良。 结果 :四种方法提取的DNA质量都能达到相关分子生物学实验的要求 ,但在操作程序、扩增效率等方面存在差异。 结论 :微波脱蜡法操作简单、快速、扩增成功率高 ,适合于应用PCR技术对病理标本进行相关研究。
Objectives: To choose a relatively ideal method for genomic DNA extraction from paraffin embeded tissues. The methods should have advantages of high DNA yied, high DNA quality, easy performance and stabilization. Methods: Four methods were used to extract DNA from paraffin embeded tissues. The results were compared in view of the advantages mentioned above. Modification was made to one of the four methods. Results: All of the four methods could give high quality of DNA, but they were different in terms of the DNA yied, time and lab input and the success rate of the PCR. Conclusions: The microwave based DNA extract method is simple, quick and reliable, so it is considered applicable to archival tissue for PCR based studies.
