醒脑益智组分中药对AD大鼠学习记忆及氧化应激-凋亡相关研究 被引量:2

Learning and memory and oxidative stress-apoptosis in Alzheimer's Disease model rats of Xingnao Yizhi multi-component Chinese Medicine Group

摘要 目的:观察醒脑益智组分中药对阿尔茨海默病(AD)模型大鼠学习记忆功能的影响及对氧化应激-凋亡相关机制的调控。方法:采用D-半乳糖(D-gal)腹腔注射联合β-淀粉样蛋白1-40(Aβ1-40)海马内注射的复合造模方法建立AD动物模型,实验分为假手术组、AD模型组、石杉碱甲0. 05 mg·kg-1组、醒脑益智组分中药179. 2、89. 6、44. 8 mg·kg-1组。以Morris水迷宫检测大鼠学习记忆能力,苏木-伊红(HE)染色观察大鼠脑组织病理学形态变化,酶联免疫吸附试验(ELisa法)检测脑组织及血清超氧化物歧化酶(SOD)活力、脂质过氧化物丙二醛(MDA)含量,免疫组化检测凋亡相关蛋白Bcl-2、Caspase-3的表达。结果:与模型组比较,各给药组大鼠逃避潜伏期降低,穿台次数增加,穿越第四象限的路程百分比也均有提高,以阳性药物和高、中剂量组作用有显著性差异(P <0. 05);各给药组能明显改善AD大鼠脑部神经元细胞形态;药物高、中剂量组能使AD大鼠脑组织及血清中SOD活性提高,MDA含量明显降低(P <0. 05),Bcl-2表达量升高,Caspase-3表达量减少。结论:醒脑益智组分中药对复合造模致AD模型大鼠学习记忆障碍具有一定改善作用,这可能与抗氧化及抗凋亡的脑保护作用有关。 OBJECTIVE To investigate the effect of Xingnao Yizhi multi-component Chinese Medicine Group on learning andmemory impairment and oxidative stress-apoptosis of Alzheimer’s Disease(AD)model rats.METHODS D-galactose intraper-itoneal injection and Aβ1-40 intrahippocampal injection were used to established the model Wistar rats,which were randomly divid-ed into sham operation group,AD model group,huperzine group 0.05 mg·kg-1,Xingnao Yizhi Multi-component Chinese Medi-cine Group at 179.2,89.6 and 44.8 mg·kg-1.Water maze test was used to evaluate the learning and memory ability of therats,hematoxylin-eosin staining to observe the pathological changes of brain tissues in rats,ELISA to test the superoxide dis-mutase(SOD)activity and lipid peroxide malondialdehyde(MDA)in brain tissue and serum,immunohistochemical method todetect the expression of apoptosis-associated protein Bcl-2 and Caspase-3.RESULTS Compared with model control group,theescape latency periods of rats in each drug group declined,the number of cross-platform increased,the percentage of journeythrough the fourth quadrant was also improved obviously in huperzine group,high and middle dose drug groups(P<0.05).Themorphology of neurons in the brain of AD rats was significantly improved in each drug group,SOD activity and Bcl-2 expressionwere increased,MDA and Caspase-3 contents were decreased significantly in Xingnao Yizhi Multi-component Chinese MedicineGroups a 179.2 mg·kg-1 and 89.6 mg·kg-1(P<0.05).CONCLUSION Xingnao Yizhi Multi-component Chinese MedicineGroup provides obvious effect on improving the dysfunction of learning and memory of the combined AD model rats,which can berelated with antioxidant and anti-apoptotic effects on the brain.
