摘要
为测定不同种类的红豆杉中紫杉醇的含量,采用ZORBAX SB-C18为分析柱,流动相为甲醇-乙腈-水=24:37:39,流速0.8mL/min,检测波长227nm。结果:紫杉醇在0.21019μg~2.10187μg(r=1.0000)范围内呈良好线性关系,平均回收率为96.4%,RSD为1.3%。结论:不同种类的红豆杉中紫杉醇的含量有一定的差异,云南红豆杉、南方红豆杉、曼地亚红豆杉、加拿大红豆杉及欧洲红豆杉中紫杉醇平均含量分别为0.034%、0.023%、0.041%、0.020%、0.024%。该次实验采用的方法简便快捷、灵敏度高、重现性好、准确可行,适用于紫杉醇原料的质量控制。
This study is aimed at determining the content of taxol in different Taxus species. ZORBAX SB-C18 column was used. The mobile phase was methanol-acetonitrile-water=24:37:39 at a flow rate of 0.8 mL/min. The detection wavelength was at 227 nm. The linear relationship of peak area was good when injection volume of Taxol was in the range of 0.21019μg-2.10187 μg(r=1.0000). The average recovery was96.4%and RSD was 1.3%. The content of taxol in different Taxus species was different, and the average taxol content in the five species were 0.034%, 0.023%, 0.041%, 0.020% and 0.024%, respectively. The method adopted in this experiment is simple, fast, sensitive, reproducible, accurate and feasible, and it is suitable for the quality control of paclitaxel raw materials.
