VEGF-C与MAT-1在三阴性乳腺癌中表达及其与预后的相关性研究 被引量:4

摘要 目的探讨VEGF与MAT-1在三阴性乳腺癌中的表达及相关性。方法采用免疫组化S-P法和Real-Time RT-PCR,分别检测三阴性乳腺癌和非三阴性乳腺癌组织中VEGF-C与MAT-1蛋白与mRNA的含量。分析其在乳腺癌发生中的作用及其与预后的相关性。结果乳腺癌组织中MAT-1 mRNA和VEGF-CmRNA的含量均高于癌旁组织,分别是4.19倍和5.27倍。MAT-1和VEGF-C蛋白在乳腺癌组织中阳性表达(62%,73%)显著高于癌旁组织(12%,18%),具有统计学差异(P<0.05)。MAT-1和VEGF-C蛋白在三阴性乳腺癌组织中阳性表达率(67%,80%)高于非三阴性乳腺癌组织(57%,67%), VEGF-C与MAT-1的表达呈正相关(r=0.217)。在不同临床分期、组织病理学分级及有/无淋巴结转移,比较MAT-1和VEGF-C蛋白阳性率,差异均有统计学意义(P<0.05)。结论 MAT-1和VEGF-C蛋白在三阴性癌组织中高表达,表达水平与临床分期、组织病理学分级和淋巴结转移密切相关,可作为判断三阴性乳腺癌预后及治疗的新靶点。 Objective To investigate the expression and correlation of MAT-1 and VEGF-C in breast cancer.Methods Immunohistochemistry S-P method and RT-PCR were used to detect MAT-1 and VEGF-C protein and mRNA levels in tiple-negative breast cancer and no tiple-negative breast cancer and their roles were analyzed in the occurrence of breast cancer and relevance with the prognosis of breast cancer.Results MAT-1 mRNA and VEGF-C mRNA in breast cancer were higher than that of breast para-carcinoma tissue,4.19 times and 5.27 times,rspectively.MAT-1 and VEGF-C protein positive expression rates in breast cancer(62%,73%)were significantly higher than breast para-carcinoma tissue(20%,18%),which was statistically significant(P<0.05).MAT-1 and VEGF-C protein positive expression rates in triple negative breast cancer(67%and 80%)were higher than that of non triple negative breast cancer(57%and 67%).MAT-1 and VEGF-C protein positive expression in breast cancer were positively correlated.MAT-1 and VEGF-C protein positive rates among different clinical stages,histological grade and lymph node metastasis were statistically significant(P<0.05).Conclusion MAT-1 and VEGF-C proteins are highly expressed in triple negative breast cancers and their expression levels correlate with clinical stage,histological grade and lymph node metastasis,which could be used as biomarkers for prognosis and new therapeutic targets in triple negative breast cancer.
