摘要
目的 :探讨复合预处理 (亚低温下缺血预处理 )对大鼠小肠缺血再灌注损伤的保护作用及机制 .方法 :将 32只大鼠随机分为 4组 (每组 8只 ) :假手术对照组 (Sham)、缺血再灌注组 (I/R)、缺血预处理组 (IP)、亚低温预处理组 (MHIP) .比较各组小肠组织湿干质量比、Ca2 + Mg2 + ATP酶含量及血清乳酸脱氢酶 (LDH)、超氧化物歧化酶 (SOD)、丙二醛 (MDA)活力、总抗氧化 (TAX)能力的变化 ,并观察各组肠组织超微结构及Bcl 2 ,Bax蛋白表达 .结果 :肠缺血再灌注后小肠湿干质量比值增高 ,LDH ,MDA含量明显上升 (P <0 .0 1 ) ,Ca2 + Mg2 + ATP酶、SOD活性及TAX能力明显下降 ,小肠组织Bcl 2和Bax蛋白表达吸光度 (A)明显增高 (P <0 .0 1 ) ;IP组与I/R组比较及MHIP组与IP组比较小肠湿干质量比值减小 ,LDH ,MDA含量明显下降 ,Ca2 + Mg2 + ATP酶、SOD活性及TAX能力明显上升 ,小肠组织Bcl 2蛋白表达吸光度 (A)值增高 ,Bax蛋白表达吸光度 (A)值降低 (P <0 .0 1 ,P <0 .0 5 ) ,Bcl 2 /Bax吸光度 (A)比值明显增高 .结论 :亚低温缺血预处理通过增加肠组织自身抗氧化能力、抑制脂质过氧化、上调Bcl
AIM: To investigate the protective effect of compound precondition(ischemic preconditioning under the mild hypothermia MHIP) on small intestine from ischemia reperfusion(I/R) injury in rats and its mechanism. METHODS: Thirty two rats were randomized into 4 groups (8 in each group): Sham operated group (sham), ischemia reperfusion (I/R) group, ischemic preconditioning (IP) group, mild hypothermia ischemic preconditioning (MHIP) group.The wet/dry ratio, Ca 2+ Mg 2+ ATPase in intestine tissue, the LDH, malondialdehyde (MDA), and activities of superoxide dismutase (SOD) and total antioxidase (TAX) in blood were compared. ultrastructure, Bcl 2 and Bax expression in intestinal mucosa tissue were observed. RESULTS: After I/R, the tissue wet/dry ratio, LDH and MDA were significantly elevated( P <0.01). Ca 2+ Mg 2+ ATPase, SOD, TAX declined obviously; the optic density of Bcl 2 and Bax proteins increased significantly( P <0.01). The tissue wet/dry ratio, LDH, MDA and the optic density of Bax protein were significantly lower in IP group than in I/R group, and in MHIP group than in IP group. The content of Ca 2+ Mg 2+ ATPase. SOD, TAX and the optic density of Bcl 2 protein were significantly higher in IP group than in I/R group ( P <0.01), and in MHIP group than in IP group ( P <0.05). CONCLUSION: MHIP can protect intestine against I/R injury in rats, which may be related to enhancing oxidation resistance of intestine, inhibiting lipid peroxidation, upregulating the protein expression of bcl 2 genes and downregulating the protein expression of bax genes and reducing myocyte apoptosis.
