兔骨髓间充质干细胞的提取与体外培养

摘要 目的 通过提取兔骨髓中间充质干细胞 (Mesenchymalstemcells,MSCs)并加以纯化 ,在体外培养条件下研究其增殖及生长特性。方法 从骨髓中提取间充质干细胞 ,体外培养扩增 ,再以相差显微镜、电镜观察并绘制生长曲线。结果 原代培养及传代培养显示 ,兔自体骨髓间充质干细胞具有活跃的增殖倍增能力 ;骨髓间充质干细胞内细胞器与其生物活性变化相一致。结论 骨髓间充质干细胞体外培养成功 ,为今后利用自体间充质干细胞通过组织工程学方法修复软骨、骨、肌肉系统的组织缺损奠定了基础。 Objective To investigate the behavior of mesenchymal stem cells in vitro. Methods MSCs (mesenchymal stem cells) were extracted from rabbit bone marrow and purified, then proliferated in vitro. They were observed under phase-contrast microscope and electronic microscope and the growth curve was drawn accordingly. Results MSCs which were in low abundance proliferated when cultured in vitro; The structures of the organellae varied with the changes of cell biological characteristics. Conclusion The success of in vitro culture of MSCs will be useful for repairment of cartilage?skeletal tissue defects by tissue engineering technique in the future.
