摘要 目的 :探讨抽动 -秽语综合征的遗传学易感因素 ,研究多巴胺D2受体启动子区域 (DRD2P)基因和儿茶酚氧位甲基转移酶 (COMT)基因在抽动 -秽语综合征发病中的作用。方法 :采用病例对照研究 ,病例组 2 4人 ,对照组 30人。DRD2P基因定型采用PCR扩增、限制性内切酶BstNⅠ消化、2 %琼脂糖凝胶电泳 ;COMT基因定型采用PCR扩增、限制性内切酶NlaⅢ消化、12 %聚丙烯酰胺凝胶电泳 ;结果用SPSS软件进行统计分析。结果 :DRD2P基因型、COMT基因型和正常对照间无显著性差异。结论 :抽动 -秽语综合征与DRD2P基因、COMT基因的多态性缺乏关联 ,仍需进一步的研究。 Objective: To examine the genetic risk factors of Tourette syndrome and explore the possible role of Dopamine D2 receptor promotor gene and Catechol-O-methyltransferase (COMT) gene in Tourette syndrome. Methods: A case control study based on hospital involving 24 Tourette syndrome patients and 30 healthy people was performed . DRD2P genotyping was carried out using polymerse chain reaction (PCR) followed by restriction enzyme BstNⅠ digestion and 2% agarose electrophoresis . COMT genotyping was carried out using polymerse chain reaction (PCR) followed by restriction enzyme NlaⅢ digestion and 12% nondenaturing polyaclamide gel electrophoresis. Statistical analysis of genotype and allele frequencies between patients and controls was performed by SPSS program . Results: No significant difference was observed between patients and controls for genotype and allele frequencies of DRD2P gene and COMT gene. Conclusion: Although the results suggested no association between DRD2P, COMT and Tourette syndrome, further studies are necessary.
