部分背根切断对备用背根节NT-3表达的影响 被引量:1

THE EFFECT OF PARTIAL DORSAL ROOT RHIZOTOMY ON THE EXPRESSION OF NT-3 IN SPARED ROOT GANGLION OF CAT

摘要 目的 探讨部分去背根后备用背根节 (L6 )各类细胞NT 3及其mRNA的含量变化。 方法 对成年雄性猫行单侧部分背根切断术 (切除一侧L1 ~L5,L7~S2 DRG ,保留L6 为备用根 )。取正常组一侧和术后 3d及 7d组手术侧的L6 DRG制作 2 0 μm厚冰冻切片 ,分别用NT 3抗体及NT 3cRNA探针行免疫组织化学及原位杂交染色。观察NT 3及其mRNA在DRG各类细胞的分布 ,测定NT 3及其mRNA在神经元和卫星细胞的光密度值 ,所得数据用q检验进行统计分析。 结果 部分去背根后 ,各时相备用背根节大神经元内NT 3的光密度值较正常者进行性减少 ,(P <0 0 5 ) ,而NT 3mRNA的光密度值术后 3d减少 ,7d回升至近正常者水平。比较之 ,小神经元和卫星细胞NT 3及其mRNA的光密度值进行性增多 (P <0 0 5 )。 结论 部分背根切断对备用背根节各类细胞NT 3表达的影响不同 ,其功能意义可能与NT Objective To explore the change of NT-3 and its mRNA expression in spared root ganglion following partial dorsal rhizotomy(L-1-L-5,L-7-S-2 DRG were sectioned,but L-6 was spared). Methods Fifteen male adult cats were used and divided into three groups.Two group animals were performed unilateral spared rhizotomy and animals were sacrificed at survived 3 days and 7 days after operation.L-6 dorsal root ganglion(DRG) from operation side and control in each animal was taken and sectioned into 20?μm thickness.Sections were stained by immunohistochemistry method(NT-3 antibody 1∶1?500, Santa cruz) and in situ hybridization technique(NT-3 cRNA probe 1∶100).The A value of NT-3 and its mRNA in the large,medium and small sized neurons and satellite cells in DRG in one side of normal animals,operation side of unilateral dorsal rhizotomy was counted and analyzed by statistic method(q test). Results The positive stains of NT-3 and its mRNA in L-6 DRG was mainly distributed in large sized neurons(>57?μm) and a few medium and small sized neurons(13-57?μm).In addition,the positive reaction was also observed in a few satellite cells.Both 3 and 7 days after partial dorsal rhizotomy,the A value of NT-3 in the large sized neurons progressively decreased(P<0^05),while that of NT-3mRNA decreased at 3 days after operation,but recovered up to normal level at 7 days after operation(P>0^05).On the contrary,the A value of NT-3 and its mRNA in the small sized neurons and satellite cells progressively increased both 3 and 7 days after operation(P<0^05).Conclusion Our results suggest that the difference on the expression of NT-3 in large and small sized neurons of DRG following partial dorsal root rhizotomy may involve in the plasticity of spinal cord.
