摘要
目的研究电针刺人中、百会穴在急性脑梗死治疗中的作用机理。方法选用短暂性大脑中动脉梗塞90min大鼠模型,运用免疫组化染色的方法,观察电针刺人中、百会对短暂性大脑中动脉梗塞90min大鼠生存因子Akt、凋亡因子caspase-9的作用。结果磷酸化Akt免疫反应增多表现在梗塞大脑半暗区的8和24h,8h为高峰期,表现在阳性细胞的数量和颜色的浓度上,活化的caspase-9激烈的表现在半暗区的8h;电针刺能抑制caspase-9的活性化,TUNEL阳性细胞减少表现在24h病灶侧大脑皮质。结论针刺能促进生存因子p-Akt增多,抑制caspase-9的活性,从而使TUNEL阳性细胞减少;针刺或许有一种潜在的减少神经细胞凋亡损伤的作用。
Objective:To investigate the effects of Eletroacupuncture (EA) on activety of ph ospho-Akt(p-Akt),cleaved caspase-9 in core and penumbra of transient midle ce rebral artery occlusion(tMCAO)in rats.Methods: After 90minutes of tMCAO in rats, to investigate the influences of EA on activety of p-Akt and cleaved caspase-9 by immunohistochemical staining.Results:p-Akt increased in the ipsilateral hem isphere after tMCAO with a peak at 8h,and EA enhanced the Akt expression both in the number and the staining strength mainlt in the ischemic penmbra (IP)at 8 an d 24h. Cleaved caspase-9 was strongly induced at 8h in IP,which was suppressed with EA. The number of terminal deoxynucleotidyl transferase-mediated uridine 5 ' triphosphate-biotin nick end labgeling (TUNEL) positive cells reduced at 24h in the cerebral cortex. Conclusion:It suggests that EA could potentiate the Akt and suppresse the caspase-9 activations,and migh have a potential to reduce the number of neuronal cells undergoing apoptotic cell death.
