摘要 目的 :观察结缔组织生长因子 (connectivetissuegrowthfactor,CTGF)mRNA在增生性玻璃体视网膜病变(PVR)增生膜 (periretinalmembranes,PRM )中的表达 ,探讨其临床意义 .方法 :采用原位杂交方法 ,对玻璃体切割手术获得的 12例PVR增生膜进行CTGFmRNA的检测 .结果 :染色阳性 9例 ,其中弱阳性 (+) 1例 ,阳性 ( ) 3例 ,强阳性 ( )5例 ,阳性率为 72 % ;阳性细胞多是一类胞体为长圆形或多角型 ,胞核呈圆形或卵圆形 ,胞质丰富的上皮样细胞 ;也有少数阳性细胞呈梭形、胞体较大的成纤维细胞样细胞 .结论 :PVR形成过程中视网膜色素上皮细胞 (RPE)、成纤维细胞在TGF β等生长因子的刺激下 ,CTGFmRNA表达显著上调 。 AIM: To investigate the expression of connective tissue growth factor (CTGF) mRNA in human periretinal membranes(PRM) of proliferative vitreoretinopathy (PVR) and to discuss its clinical significance. METHODS: In situ hybridization was used to detect the presence of CTGF mRNA in twelve periretinal membranes of PVR which were acquired by vitrectomy. RESULTS: CTGF mRNA expression was detected in four PVR C grade and five D grade membranes and CTGF mRNA expression was found to be located in cytoplasm of all positive cells. Among them, one membrane showed weak positive staining (+), three membranes showed positive staining () and the remaining five membranes showed strong positive staining (). The average positive rate was 72%. Positive staining cells were seen as a type of epithelial like cells and fibroblast like cells. CONCLUSION: CTGF mRNA is highly expressed and up regulated in PRM by cytokine stimulation and it may play an important role in the pathogenesis of PVR.
