水稻蜡质基因(Wx)反义片段在转基因后代中的遗传稳定性 被引量:4

摘要 用根癌农杆菌 (Agrobacteriumtumefaciens) 介导的遗传转化方法 ,将携带水稻蜡质基因反义片段导入中花 11号粳稻中 ,分析了 2 0个独立转化的转基因植株。经GUS染色、PCR分子检测 ,证明蜡质基因反义片段已整合到这些植株的染色体组中 ;在转基因植株后代T1的籽粒外观糯性检测中 ,有 8个转基因植株检测到糯性籽粒的存在 ;在 12个转基因植株T2后代直链淀粉含量测定中 ,有 5个T3的直链淀粉含量比对照有明显的降低 ;在T3、T4的直链淀粉含量测定中 ,获得了直链淀粉含量比对照低 1%的株系 ;来自T2的糯稻株系其后代能相对稳定遗传。在籽粒GUS检测和外观糯性检测中 。 Through the genetic transformation mediated by Agrobacterium tumefaciens, an antisense waxy gene was inducted into in Zhonghua No.11 (Oryza sativa L. ssp. japonica cv.), and 20 transgenic linestransformed independently were analyzed. The assay of GUS and PCR indicated that an antisense waxy gene segment had been integrated into the genome of those lines. The detection of appearance waxiness of T1 seeds of the transgenic plants found that there were waxy seeds in 8 transgenic plants. The test of amylose content of T3 seeds of 12 transgenic plants showed that 5 out of 12 had a marked decrease. The lines whose seeds's amylose contents were 1% lower than that of non-transgenic plants were selected from T3 and T4 plants through the content assay. The appearance waxiness of seeds from waxy rice T2 plants could stably inherit. In addition, the assay of GUS and appearance waxiness showed that there were differences between segregation ratio of GUS in endosperm and segregation ratio of waxy and non-waxy kernels.
