人巨细胞病毒感染对单核细胞和内皮细胞株ECV-304间趋化和黏附的增强作用 被引量:1

摘要 目的 研究人巨细胞病毒(HCMV)感染对单核细胞及内皮细胞株表达趋化因子的影响,以及对两者之间趋化和黏附的影响。方法 通过ELISA法和半定量RT-PCR的方法,检测CMV感染对单核细胞株(THP-1)及内皮细胞株(ECV-304)几种趋化因子及其受体表达的改变,同时通过趋化实验和黏附实验研究内皮细胞对单核细胞的趋化和黏附的改变。结果 HCMV感染可刺激ECV-304分泌MCP-1、GROα,在mRNA水平可改变内皮细胞MCP-1、CRO-α和IL-8表达,先下调后上调(IL-8除外);而对单核细胞趋化因子受体CCR2、CXCR2的表达在mRNA水平却有明显的下调作用(P<0.05)。HCMV感染可显著增强内皮细胞对单核细胞的趋化和黏附。结论 HCMV感染可增强内皮细胞对单核细胞的趋化和黏附,参与炎症反应,这可能与CMV上调内皮细胞趋化因子表达有关。 Purpose: To study the effect of human cytomegalovirus (HCMV) infection on the expression of chemokines and their receptors of monocytes and endothelial cells and on the chemotaxis and adhesion of endothelial cells to monocytes. Methods: The expression of chemokines and chemokine receptors in endothelial cells ECV-304 and monocytes THP-1 was studied by using ELISA and semi-quantitative RT-PCR. The chemoattract test and adhesion test were used to detected the functional alteration of endothelial cells on monocytes. Results: After infection, the secretion of MCP-1, GROα by ECV-304 was enhanced, the expression of MCP-1, GRO-α of endothelial cells was down-regulated and then up-regulated in mRNA level, the expression of chemokine receptors CCR2 and CXCR2 (P < 0.05) was down-regulated in THP-1 cell lines, the chemoattract and adhesion of endothelial cell to monocytes were enhanced. Conclusions: CMV infection enhanced the chemoattract and adhesion of endothelial cell to monocytes, which maybe mediate by regulation of expression of chemokines and chemokine receptors of endothelial cells and monocytes.
