摘要
目的:建立同时测定复方肉苁蓉合剂中毛蕊花糖苷、党参炔苷和丹酚酸B的HPLC方法。方法:色谱柱:Inertsil C18柱(250 mm×4.6 mm,5μm),流动相(乙腈-0.1%磷酸水溶液=19∶81);流速1.0 m L/min;柱温35℃;毛蕊花糖苷、党参炔苷和丹酚酸B检测波长分别为330、286、268 nm。结果:毛蕊花糖苷、党参炔苷和丹酚酸B分别在11.2~112、5.06~50.6、15.6~156μg/m L范围内与色谱峰峰面积线性关系较好,相关系数分别为0.9998、0. 9997、0.9998,加样回收率的平均值分别为99.11%、97.81%、98.30%,RSD分别为1.94%、2.03%、1.35%(n=6)。结论:所建立的HPLC含量测定方法快速、准确,可同时测定复方肉苁蓉合剂中毛蕊花糖苷、丹酚酸B和党参炔苷的含量,为复方肉苁蓉合剂的质量分析与控制提供一定的参考。
Objective To establish a HPLC method for simultaneously determining verbascoside,codonopsis and salvianolic acid B in Fufang Roucongrong mixture.Methods HPLC method was performed on a Inertsil C18 column(250 mm×4.6 mm,5μm)with mobile phase(acetonitrile-0.1%phosphoric acid solution=19∶81),flow rate 1.0 mL/min and column temperature 35℃.The detection wavelengths of of verbascoside,codonopsis glycoside and salvianolic acid B were 330,286,268 nm respectively.Results Verbascoside,codonopsis glycoside and salvianolic acid B in the corresponded ranges of 11.2~112,5.06~50.6,15.6~156μg/mL demonstrated good linear relationship with chromatographic peak area.The correlation coefficients were 0.9998,0.9997 and 0.9998,the average recoveries were 99.11%,97.81%and 98.30%,and RSDs were 1.94%,2.03%and 1.35%(n=6),respectively.Conclusion The established HPLC method is rapid and accurate.It can simultaneously determine the content of verbascoside,codonopsis glycoside and salvianolic acid in Fufang Roucongrong mixture and provide a reference for the control and quality analysis of the compound.
