干扰素β-1b对多发性硬化症末梢血细胞因子变化的影响

摘要 目的 探讨多发性硬化症 (Multiplesclerosis ;MS)患者使用干扰素 β 1b(Interferonbeta - 1b ,IFNβ 1b)治疗前后末梢血中细胞因子的变化过程及对其平衡环境的影响。方法  13例患者用药前两次及用药后第 1、2、4、12周分别进行检测末梢血细胞因子 ,采用肝素加末梢血经 4hPMA2 5ng/ml及Ionomycin 1μg/ml刺激培养后 ,用IFN r、IL 4、IL 5以及IL 10的单克隆抗体分别对CD4及CD8阳性的T细胞内的细胞因子进行染色 ,再用微量测定法(Phrocytomotoly ,PCM)进行测定。结果 Th1细胞因子用药前后无任何变化。Th2细胞因子在用药第 4周会一过性上升 ;在 12周以后又回归到用药前水平。另外 ,IFN r/IL 4的比率也在用药 4周后开始降低逐渐又恢复到用药前水平。结论 干扰素 β 1b对末梢血中细胞因子的影响有其时间依赖性 ,通过免疫调节作用可以抑制MS疾病的发展。 Objective To study cytokine in peripheral blood of the patients with multiple sclerosis(MS)and how it effects balance state at particular time before and after therapy with IFNβ 1b.Methods The CD4+ and CD8+ T cells were stained with monoantibody to IFNr?IL 4?IL 5?IL 10 after stimulated culture for 4 hours.Compare analysis of two assay before the therapy and assays at 1,2,4,12 weeks after therapy was performed in 13 cases respectly.Results No change of Th1 cytokine before and after therapy was detected and Fugacious rise in fourth week after theraphy with cytokine,came back to primary level after 12 weeks additionally,IFNr/IL 4 gradually decreased in 4 weeks after therapy to primary level.Conclution The IFNβ 1b effect on peripheral blood cytokine relies on variable time.Therefore,immune regulation may inhabit development of MS.
