摘要
目的 :观察携带人肝细胞生长因子基因的重组腺病毒 (Ad_HGF)对大鼠局灶性脑缺血的治疗作用。方法 :建立大鼠局灶性脑缺血动物模型。携带人肝细胞生长因子基因的穿梭质粒与携带复制缺陷的人血清 5型腺病毒基因组的质粒在 2 93细胞进行同源重组 ,构建Ad_HGF。携带绿色荧光蛋白基因的重组腺病毒 (Ad_GFP)载体注射到局灶性缺血大鼠的缺血侧侧脑室 ,观察其在脑内的表达部位和持续时间。动物给药治疗后 3d ,取大脑 ,平均切成 4片 ,用TTC染色评价Ad_HGF对缺血的治疗效果。结果 :Ad_GFP仅在大鼠双侧侧脑室壁及脑室内的脉络膜细胞表达 ,表达持续两周左右 ;Ad_HGF治疗组动物大脑缺血区明显小于非治疗组动物 (P <0 .0 5 )。结论 :Ad_HGF能显著缩小大鼠局灶性脑缺血区。
Objective:To investigate the therapeutic effect of adenovirus-mediated expression of hepatocyte growth factor gene(HGF) on transient focal cerebral ischemia in a rat modal.Methods:A rat model of transient focal cerebral ischemia due to middle cerebral artery(MCA) occlusion was adopted.Recombinant adenovirus carrying HGF gene(Ad-HGF) was constructed in 293 cells.After intraventricular injection of adenovirus carrying green fluorescent protein(Ad-GFP),a reporter gene,the expression was observed.After single cerebral ventricular injection of Ad-HGF into rats of the treatment group immediately after surgery,its therapeutic effect on transient focal cerebral ischemia was evaluated by TTC staining of the frontally sectioned brain removed on day 3.Result:GFP expression was visualized only in the ependymal cells and cells surrounding ventricular region in both hemispheres from day 1 to day 14.In rats of the treatment group,the infarct volume was significantly smaller after application of HGF compared with control rats without injection of Ad-HGF(P<0.05).Conclusion:Ad-HGF reduced the infarct areas after transient focal cerebral ischemia in rats.
