摘要
用改良的叶圆盘法进行了烟草bar基因高效转化并获得 11个转基因株系 ,转化率高达 32 .4 %。提出构建以bar基因为选择标记的新型植物表达载体的设想 ,实验分析了不同PPT浓度与GS酶的解抑制量及外植体存活、分化率的关系 ,结果表明 ,10 0mg L以上的PPT可完全抑制非转化外植体的存活 ,从而证实了设想的可行性。研制出检测抗除草剂转基因植物的微量、快速、有效的检测新方法 ,并用实际检测实验和统计学手段验证了该方法的可靠性和实用性。
Using modified leaf-disk method, a tobacco bar gene transformation has been conducted and 11 transgenic plants were gained. The transformation efficiency reached 32.4%.A new tentative idea for constructing a new plant expressible plasmid with bar gene as the selective marker has been postulated. Also, the relationship between PPT concentrations, decreases of inhibition to the GS enzyme activity and survival rate and differentiation efficiency of explants has been analyzed. The results state that 100 mg/L or much higher concentrations of PPT can inhabit the vigor of explants thoroughly, which confirm the feasibility for the new idea. A new test method has been developed that is suitable for the rapid and effective assay of the mini-trans-bar-genic bud or explant, whose practicability and credibility has been validated by the means of practical test experiments and statistic analysis.
