hMAMmRNA及其蛋白在乳腺癌组织中的表达和临床意义 被引量:3

Expression and Clinical Significance of Human Mammaglobin mRNA and Its Protein in Breast Cancer Tissue

摘要 目的检测乳腺癌组织中人乳腺球蛋白(h MAM)mRNA及其蛋白的表达,验证h MAMmRNA与蛋白表达是否一致,探讨其表达与乳腺癌发生、发展的关系。方法采用实时荧光定量PCR(FQ-PCR)法检测30例乳腺癌组织及其相应癌旁正常组织,15例乳腺良性病变组织中h MAMmRNA的表达,采用组织微阵列技术(TMA)通过免疫组化法(IHC)检测116例乳腺癌组织,44例癌旁正常组织中及32例乳腺良性病变组织中h MAM蛋白的表达情况。结果 h MAMmRNA与其蛋白在乳腺癌组织表达量(率)分别为(2.73±0.55)和59.48%,高于癌旁正常组织及乳腺良性病变组织中的表达(P均<0.01);h MAMmRNA及其蛋白在乳腺癌组织中阳性表达的一致率为83.33%(Kappa=0.453,P<0.01);h MAM蛋白表达与乳腺癌腋窝淋巴结转移、组织学分级、ER有关,即腋窝淋巴结转移者、组织学II-III级、ER阳性者h MAM蛋白阳性表达率高于无转移者、组织学I级、ER阴性者(P均<0.05);Logistic回归分析表明乳腺癌组织中h MAM表达与组织学分级、ER、PR均相关(P均<0.05)。结论 h MAMmRNA及其蛋白特异性高表达于乳腺癌组织,有望成为判断乳腺癌恶性程度及侵袭能力的独立指标。 Objective To investigate the expression and clinical significance of the human mammaglobin( h MAM) mRNA and its protein in breast cancer tissue,to verify consistency of their expression in breast cancer,and ti analyze their relationships with the development of breast cancer. Methods The expression of h MAMmRNA was determinated by FQ- PCR method in breast tissues and the adjacent normal tissues of 30 breast cancer cases and 15 benign gastric lesion tissues. The expression of h MAM protein was determined by using IHC in 146 breast cancers tissues,74 adjacent normal breast tissues and 47 benign breast lesion tissues.Results The relative quantity of h MAMmRNA and the positive rate of h MAM protein in breast tissues were( 2. 73 ± 0. 55) and 59. 48%,which were significantly higher than those in adjacent normal tissues and benign breast lesion tissues( P < 0. 01). The agreement rate of h MAMmRNA and its protein in breast cancer tissues was 83. 33%( Kappa value was 0. 453,P < 0. 01). The expression of h MAM protein was significantly different in different histologic grade,groups with or without lymph node metastasis and estrogen receptors. The expression rates of h MAM protein in group with lymph node metastasis,the histologic grade of II and III and the one positive estrogen receptor were signficantly higher than those without lymph node metastasis,the histologic grade of I and negative estrogen receptor( P all < 0. 05). Multivariate logistic regression analysis showed that the expression of h MAM had relationship with histologic grade,estrogen receptor and progesterone receptor( P all < 0. 05). Conclusion h MAMmRNA and its protein was highly expressed in breast cancer tissues.Therefore,the expression of h MAMmRNA or its protein was take as a independent indicators of malignancy and invasion for breast cancer in clinical application.
