缺牙对幼年大鼠海马CA1区神经元凋亡及casepase-3蛋白表达的研究

摘要 目的通过不同缺牙数目幼年大鼠海马CA1区神经元细胞凋亡及海马神经元中天冬氨酸特异性半胱氨酸蛋白酶(Aspartate-specific cysteine protease-3 casepase-3)的表达情况,探讨幼年大鼠缺牙后海马神经细胞凋亡可能的机制。方法 50只雄性SD大鼠,4-5周龄,随机分成A组空白对照组,B组3颗磨牙缺失组,C组6颗磨牙缺失组,D组9颗磨牙缺失组,E组12颗磨牙缺失组。实验采用HE染色、免疫组化法检测casepase-3蛋白的表达情况,并用末端标记(TUNEL)法观察大鼠海马组织CA1区神经细胞凋亡的变化。结果随着缺牙数目增多,幼年大鼠海马CA1区细胞结构可见肿胀、空泡、结构排列紊乱、神经元细胞显著减少等明显组织学改变。2.个别磨牙缺失可引起海马神经元细胞组织学结构轻微变化及casepase-3表达轻度增加,但与对照组相比无统计学差异(P>0.05)。3.缺牙4周后,多颗磨牙缺失组幼年大鼠海马CA1区细胞的凋亡细胞增多,casepase-3表达增加,与对照组相比具有统计学差异(P<0.01)。结论多数磨牙缺失会导致幼年大鼠海马区细胞凋亡增多,同时出现casepase-3蛋白的表达增加,提示多个磨牙缺失可能会促使幼年大鼠海马组织细胞凋亡。 Objective To investigate the possible mechanism underlying the apoptosis of hippocampal neurons in juvenile rats after tooth loss by studying the apoptosis of neurons in the hippocampal CA1 region and the expression of aspartate-specific cysteine protease-3 casepase-3 in the hippocampal neurons in juvenile rats that lost different numbers of teeth. Methods Fifty male Sprague-Dawley rats aged 4~5 weeks were randomly divided into the blank control group(group A), the 3-molar absence group(group B), the 6-molar absence group(group C), the 9-molar absence group(group D), and the 12-molar absence group(group E). HE staining and immunohistochemistry were used to detect the expression of casepase-3 protein, and the apoptosis of neurons in the hippocampal CA1 region was observed by terminal deoxynucleotidyl transferase dUTP nick end labeling(TUNEL). Results(1)With the increase in the number of missing teeth, the cell structure in the hippocampal CA1 region of juvenile rats showed significant histological changes such as swelling, vacuoles, disordered structure, and significant decrease of neurons.(2) The loss of some individual molars can cause slight changes in the histological structure of hippocampal neurons and a slight increase in casepase-3 expression,but there is no statistically significant difference compared with the control group(P>0.05).(3)At 4 weeks after tooth loss, the apoptotic cells increased in the hippocampal CA1 region of the juvenile rats in the groups with absence of multiple molars, and the expression of casepase-3 increased, with statistically significant difference from the control group(P<0.01). Conclusion Loss of multiple molars will lead to increased apoptosis in the hippocampal region of juvenile rats, and the expression of casepase-3 protein will increase, suggesting that loss of multiple molars may promote apoptosis of hippocampal neurons in juvenile rats.
