摘要
目的探讨清肺方对博莱霉素诱导的间质性肺炎大鼠的作用及其作用机制。方法选择清洁级SD大鼠48只,按照随机数字表法抽出8只作为正常组,其余大鼠通过气管内灌注博来霉素方法建立间质性肺炎模型。将造模后大鼠随机分为模型组、地塞米松组、清肺方高剂量组、清肺方中剂量组、清肺方低剂量组,每组8只。造模24 h后,清肺方高、中、低剂量组分别给予浓度为4.16 g/mL、2.08 g/mL、1.04 g/mL清肺方水煎液1 mL/100 g灌胃,均1次/d;地塞米松组给予浓度为0.1 mg/mL的地塞米松溶液0.875 mg/kg灌胃,每周2次;模型组及正常组均给予生理盐水1 mL/100 g灌胃,1次/d。各组均连续灌胃28 d。观察各组大鼠肺组织病理表现,检测肺组织中羟脯氨酸(HYP)、转化生长因子-β_1(TGF-β_1)、肿瘤坏死因子-α(TNF-α)水平。结果清肺方高、中、低剂量组大鼠肺组织纤维化程度均明显轻于模型组;模型组大鼠肺组织中HYP、TGF-β_1、TNF-α水平均明显高于正常组(P均<0.05),清肺方高、中、低剂量组肺组织中HYP、TGF-β_1、TNF-α水平均明显低于模型组(P均<0.05)。结论清肺方可明显减轻博来霉素诱导的间质性肺炎大鼠肺组织纤维化程度,机制可能与其抑制肺组织中TGF-β_1、TNF-α表达有关。
Objective It is to investigate the effect of Qingfei decoction on bleomycin-induced interstitial pneumonia in rats and its mechanism of action. Methods A total of 48 SD rats of clean grade were selected in which 8 rats were randomly selected as normal group according to the random number table method. The other rats were given intratracheal infusion of bleomycin to establish interstitial pheumonia, and were randomly divided into model group, dexamethasone group, Qingfei decoction high dose group, Qingfei decoction middle dose group and Qingfei decoction low dose group, with 8 rats in each group. After modeling for 24 hours, Qingfei decoction high, medium and low dose groups were given a concentration of 4.16 g/mL, 2.08 g/mL, 1.04 g/mL Qingfei decoction 1 mL/100 g, 1 time/d;dexamethasone group was given a concentration of 0.1 mg/mL dexamethasone solution 0.875 mg/kg by gavage twice a week;model group and normal group were given normal saline 1 mL/100 g by gavage, once/d. Each group was continuously intragastrically administered for 28 days. The pathological changes of lung tissues in each group were observed, and the levels of hydroxyproline(HYP), TGF-β1 and TNF-α in lung tissues were detected. Results The degree of pulmonary fibrosis in the high, middle and low dose groups of Qingfei decoction was significantly better than that in the model group. The levels of HYP, TGF-β1 and TNF-α in the lung tissue of the model group were significantly higher than those in the normal group(P<0.05), the levels of HYP, TGF-β1 and TNF-α in lung tissue of Qingfei decoctioin high, middle and low dose groups were significantly lower than those in the model group(P<0.05). Conclusion Qingfei decoction can significantly reduce the degree of pulmonary fibrosis in rats with bleomycin-induced interstitial pneumonia. The mechanism may be related to the inhibition of TGF-β1 and TNF-α expression in lung tissue.
